摘　　要：目的:通过网络药理学研究探索中药黄芪治疗免疫球蛋白A型肾病(Immunoglobulin A Nephropathy,IgAN)的潜在靶点及作用机制。方法:使用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)筛选黄芪的有效成分和靶点,通过GeneCards、DisGeNET、TTD、DrugBank、GAD等数据库查找与IgAN相关的靶点。利用Cytoscape软件,分析黄芪治疗IgAN的作用靶点和有效成分之间的关系网络,通过DAVID数据库分析黄芪治疗IgAN的有效靶点的生物学功能和作用机制。结果:根据口服生物利用度和药物相似性原则筛选出黄芪有效成分20个,筛选出的有效成分作用靶点有180个。在不同数据库中筛选出有关IgAN靶点1267个。基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析获得一系列生物功能和信号通路,如晚期糖基化终末产物(Advanced Glycation End Products,AGEs)-AGE受体(Receptor of AGE,RAGE)信号通路等。结论:黄芪通过炎症因子、血管内皮生长因子、蛋白激酶等作用于多条信号通路来实现多靶点、多通路治疗IgAN的目的。AGEs-RAGE信号通路是黄芪治疗IgAN的重要通路。黄芪可能直接或间接参与调节炎症、细胞凋亡等作用机制来治疗IgAN。Objective:To explore the potential targets and action mechanism of Huangqi(astragalus)on IgAN through network pharmacology.Methods:The effective components and targets of Huangqi was screened by TCMSP database.The targets related to IgAN were searched through GeneCards,DisGeNET,TTD,DrugBank,GAD and other databases.Cytoscape software was used to analyze the relationship between the effective target of and active components of Huangqi in the treatment of IgAN.The biological function and action mechanism of the effective targets of Huangqi in the treatment of IgAN were analyzed by DAVID database.Results:According to the principles of oral bioavailability and drug-likeness,20 active ingredients of Huangqi were screened,with 180 targets of active ingredients.A total of 1267 targets related to IgAN were screened in different databases.The GO enrichment analysis and KEGG pathway enrichment analysis gained a series of biological functions and signaling pathways,such as AGEs-RAGE signaling and others.Conclusion:Huangqi treats IgAN through multiple targets and multiple pathways.Its effective targets mainly act on multiple signaling pathways through inflammatory factors,vascular endothelial growth factor,protein kinases and so on.The AGEs-RAGE signaling pathway is an important pathway for Huangqi to treat IgAN.Huangqi may directly or indirectly participate in the regulation of inflammation,apoptosis and other mechanisms to treat IgAN.
