加味泻青丸对多发性抽动症模型大鼠行为学及肠道菌群的调节作用

摘　　要：目的:观察加味泻青丸对多发性抽动症大鼠模型行为学及肠道菌群的影响。方法:将SD雄性大鼠40只随机分为空白组10只,造模组30只,采用亚氨基二丙腈腹腔注射造模,建立多发性抽动症动物模型,将造模成功的大鼠随机分为三组:模型组、泰必利组、加味泻青丸组,每组10只。从造模成功之日起开始灌胃给药,空白组与模型组给予蒸馏水,其余组给予相应药液。2周给药结束后,观察用药前后大鼠的行为学改变情况,16S rDNA测序检测肠内容物的菌群构成。结果:用药后,加味泻青丸组大鼠的刻板行为和运动行为评分显著降低(P<0.05);加味泻青丸组Ace指数和Chao1指数升高;加味泻青丸组拟杆菌门比例降低;泰必利组、加味泻青丸组乳酸杆菌属比例升高,差异均具有统计学意义(P<0.05)。结论:加味泻青丸可显著减少多发性抽动症大鼠模型的运动及刻板行为,其机制可能与改善肠道菌群构成相关。Objective:To observe the influences of Xieqing Wan(泻青丸)on ethology and intestinal flora of tourette syndrome rats model.Methods:40 SD male rats were randomly divided into the blank group of 10 rats,and the molding group of 30 rats.Intraperitoneal injection of iminodipropionitrile was used to establish the animal model of tourette syndrome.The successful model rats were randomly divided into 3 groups:the model group,the taipiride group,and the Xieqing Wan group,with 10 rats in each group.From the date when the model was successfully established,the rats were given gavage,the blank group and the model group were given distilled water,and the other groups were given the corresponding liquid.Results:After medication,the stereotyped behavior and motor behavior scores in the Xieqing Wan group were decreased significantly(P<0.05),and regulation effect in the Xieqing Wan group was more obvious than that in the taipiride group.The Ace index and Chaol index in the Xieqing Wan group were increased.The proportion of bacteroidetes in the Xieqing Wan group was decreased.The proportion of lactobacillus genus in the taipiride group and the Xieqing Wan group was increased,and the difference was statistically significant(P<0.05).Conclusion:Xieqing Wan can significantly reduce the motor and stereotyped behavior of tourette syndrome,and its mechanism may be related to the improvement of the composition of intestinal flora.
