摘  要:器官大小是植物重要的形态性状之一。为解析棉花GhSAP在调控器官大小方面的功能及机制,我们对该基因的基本生物学特性、亚细胞定位及其互作蛋白进行了系统分析。结果显示,GhSAP含有3个WD40 repeat功能区,具有两个外显子和一个内含子。亚细胞定位结果显示,GhSAP-GFP荧光信号与DAPI共定位在细胞核上。利用酵母双杂交技术共筛选到40个GhSAP的互作蛋白,结合转录组测序结果和基因功能分析,发现Gh_D06G0323(GhAUF1)与GhSAP共表达,推测GhAUF1可能与GhSAP互作共同调控棉花器官的发育。本研究对GhSAP基因进行了深入分析,为进一步揭示植物器官大小调控机制提供了依据。Plant organ size is one of the most important morphological traits in plants.To analyze the function of Gh SAP and the underlying mechanisms of organ size control in cotton,the biological characteristics,subcellular localization and possible interacting proteins of this gene were systematically investigated.The results showed that GhSAP contains three WD 40 repeat domains,as well as two exons and one intron.Subcellular localization analysis showed that GhSAP-GFP fluorescence signal co-located with DAPI in the nucleus.A total of 40 GhSAP-interacting proteins were identified by Yeast two-hybrid system.Combined with transcriptome sequencing results and gene function analysis,it was found that Gh_D06G0323(GhAUF1)was co-expressed with GhSAP,suggesting that GhAUF1may interact with GhSAP to jointly regulate cotton organ development.In the present study,GhSAP was cloned and analyzed in detail,providing a basis for further revealing the molecular mechanism of organ size control in plants.
