摘 要:通过中药系统药理学数据库与分析平台(TCMSP)查询淫羊藿主要活性化合物;利用CTD、GeneCards和HIT等数据库收集淫羊藿主要活性化合物的作用靶点和免疫耐受的疾病靶点;通过STRING数据库构建蛋白-蛋白相互作用网络,并利用Cytoscape软件对网络中的节点进行MCODE聚类分析和网络拓扑学分析;利用DAVID数据库分析靶点的基因功能及信号通路,以探讨淫羊藿调节免疫耐受的作用机制;通过MOE 2020软件将淫羊藿主要活性化合物与其调节免疫耐受的潜在核心作用靶点进行分子对接。结果表明:淫羊藿含有淫羊藿苷、谷甾醇和山奈酚等23个主要活性成分,淫羊藿调节免疫耐受的关键靶点包括IL-6、STAT3、CXCL8等,核心通路主要为IL-STAT信号通路、FoxO信号通路和JAK-STAT信号通路等。这一研究说明淫羊藿的淫羊藿苷、谷甾醇和山奈酚等主要活性成分可作用于IL-6、STAT3、CXCL8等靶点,激活或抑制IL-STAT等相关信号通路调节机体免疫耐受状态。The main active compounds of Epimedii Folium were searched from TCMSP.CTD,GeneCards and HIT databases were used to collect the targets of the main active compounds of Epimedii Folium and the disease targets of immune tolerance.The protein-protein interaction network of compound-disease targets was constructed by STRING database,and the network were analyzed by MCODE cluster analysis and network topology analysis.DAVID database was used for the enrichment of the gene function and signaling pathway of targets to explore the mechanism of Epimedii Folium regulating immune tolerance.Futhermore,the main active compounds of Epimedii Folium were docked with the potential core targets for regulating immune tolerance by MOE 2020 software.The results showed that Epimedii Folium contained of 23 main active compounds,including icariin,sitosterol and kaempferol,etc.The key targets of Epimedii Folium regulating immune tolerance included IL-6,STAT3 and CXCL8,and the core pathways were mainly IL-STAT signaling pathway,FoxO signaling pathway and JAK-STAT signaling pathway.The results indicated that the main active compounds of Epimedii Folium such as icariin,sitosterol and kaempferol could act on IL-6,STAT3,CXCL8 and other targets,activate or inhibit IL-STAT and other related signaling pathways to regulate immune tolerance.
