半枝莲-白花蛇舌草药配伍对乳腺癌肺转移及细胞增殖的影响研究

摘　　要：目的:分析半枝莲-白花蛇舌草药配伍治疗对乳腺癌肺转移及细胞增殖的影响。方法:分别进行人高转移性乳腺癌细胞株(MDA231-LM2)及人乳腺癌细胞(MDA-MB-231)的培养,观察1组注入半枝莲-白花蛇舌草溶液、观察2组注入半枝莲溶液、对照组注入生理盐水,以荧光PCR法检测各组细胞白介素-6(IL-6)、基质金属蛋白酶1(MMP1)和趋化因子CXC基序配体1(CXCL1)表达情况,并进行比较;以酶联免疫法检测滴药12h、24h及48h后细胞570 nm吸收值,进而分析细胞增殖抑制率。结果:观察组各组细胞中IL-6、CXCL1、MMP1水平均低于对照组,且观察1组低于观察2组(P<0.05)。观察1组、2组于12 h、24 h及48 h细胞各时段吸收值均小于对照组,且观察1组各时段吸收值明显小于观察2组(P<0.05);随着滴药时间推移,观察1组抑制率逐渐升高。结论:半枝莲-白花蛇舌草药配伍后对乳腺癌肺转移有积极控制效果,对细胞增殖产生明显的抑制效果。Objective:To analyze the effects of herbal compatibility of Scutellaria barbata and Hedyotis diffusa on lung metastasis and cell proliferation of breast cancer.Methods:Human highly metastatic breast cancer cell line(MDA231-LM2)and human breast cancer cell line(MDA-MB-231)were cultured respectively.The observation group 1 was injected with Scutellaria barbata-Hedyotis diffusa solution,the observation group 2 was injected with Scutellaria barbata solution and the control group was injected with normal saline.The expressions of interleukin-6(IL-6),matrix metalloproteinase 1(MMP1)and chemokine CXC motif ligand 1(CXCL1)were detected by fluorescence PCR in each group and were compared.The 570 nm absorption values of the cells after 12 h,24 h and 48 h of instillation were detected by enzyme-linked immunosorbent assay,and then the inhibition rate of cell proliferation was analyzed.Results:The levels of IL-6,CXCL1 and MMP1 in the cells of the observation groups were lower than those of the control group,and the levels in the observation group 1 were lower than those in the observation group 2(P<0.05).The cell absorption values in the observation group 1 and group 2 at 12 h,24 h and 48 h were lower than those in the control group,and the absorption value at each time period was significantly lower in the observation group 1 than that in the observation group 2(P<0.05).With the passage of time,the inhibition rate was gradually increased in the observation group 1.Conclusion:The herbal compatibility of Scutellaria barbata and Hedyotis diffusa has a positive control effect on lung metastasis of breast cancer,and has a significant inhibitory effect on cell proliferation.
