摘 要:【目的】对大豆(Glycine max)全基因组BBX(B-box)蛋白基因进行鉴定和生物信息学分析,探究其对非生物逆境胁迫的响应模式,为大豆BBX基因的应用提供参考。【方法】利用生物信息学方法鉴定大豆BBX基因家族成员,分析其保守结构域、系统进化树、基因复制关系、顺式作用元件、组织特异性表达和非生物逆境胁迫(干旱和100 mmol/L NaCl胁迫,均处理0,1,6,12 h)响应模式。【结果】大豆BBX基因家族(GmBBXs)有42个成员,分布在除2和16号染色体外的18条染色体上。保守结构域分析表明,GmBBXs在N端有一个或两个B-box结构域(B-box1和B-box2),B-box1较B-box2保守;部分成员在C端存在一个CCT结构域。系统发育分析表明,GmBBXs家族成员分为5个亚家族,分别为B1+B2+CCT(Ⅰ型和Ⅱ型)、B1+CCT(Ⅲ型)、B1+B2(Ⅳ型)和B1(Ⅴ型)类型,其成员数量分别为3,8,6,18和7。共线性分析发现,在GmBBXs基因的扩展中存在串联复制和片段复制事件,且片段复制事件比例较串联复制大。GmBBXs基因启动子上含有多种顺式作用元件,如光应答元件、逆境胁迫应答元件和激素响应元件等。GmBBXs基因表达分析结果显示,GmBBXs在大豆根、茎、叶、花、荚果和种子中的表达存在差异,不同GmBBXs响应干旱和盐胁迫的方式不同,获得的36个GmBBXs基因表达数据可分为6种表达模式。【结论】在大豆中共鉴定得到42个GmBBXs基因,分布于18条染色体上,其结构域较为保守;GmBBXs的表达具有组织特异性,参与干旱和盐胁迫响应。【Objective】This study conducted gene identification and bioinformatics analysis on the BBX(B-box)protein of soybean(Glycine max)and analyzed the response mode to abiotic stress to provided reference for the application of soybean BBX genes.【Method】Based on the whole genome sequence of soybean,bioinformatics methods were used to identify BBX genes and analyze the conservative domains,phylogenetic tree,gene duplication,cis-acting elements,tissue-specific expression and expression pattern under abiotic stresses of drought and 100 mmol/L NaCl for 0,1,6 and 12 h.【Result】There were 42 BBX family proteins in soybean and the GmBBXs genes distributed on 18 chromosomes except chromosomes 2 and 16.Conserved domain analysis showed that GmBBXs had one or two B-box domains(B-box1 and B-box2)at the N-terminus and some members had the CCT domain at the C-terminus.B-box1 was more conserved than B-box2.Phylogenetic tree analysis showed that GmBBXs were divided into five subfamilies of B1+B2+CCT(TypeⅠand TypeⅡ),B1+CCT(TypeⅢ),B1+B2(TypeⅣ)and B1(TypeⅤ)with numbers of 3,8,6,18 and 7,respectively.Tandem replication and segmental replication events were found in the expansion of GmBBXs genes by collinearity analysis and the ratio of segmental replication events was larger than that of tandem replication.The promoter sequence of GmBBXs genes contained multiple cis-acting elements such as light response elements,stress response elements and hormone response elements.Further analyses showed that the expression abundance of GmBBXs was different in root,stem,leaf,flower,pod and seed.Transcriptome data analysis found that different GmBBXs responded to drought and salt stress in different ways.The obtained 36 GmBBXs had six different expression patterns.【Conclusion】There were 42 GmBBXs genes identified in the whole genome of soybean on 18 chromosomes with conserved domains.The expression of GmBBXs was tissue-specific and involved in drought and salt stress responses.
