摘 要:目的研究大鼠C6胶质瘤模型注射一款新型超小超顺磁性氧化铁(USPIO)纳米颗粒后的磁共振强化效果随不同时间、给药体积、浓度的变化规律,探讨合适的增强扫描注射方案。资料与方法将25只大鼠胶质瘤模型随机分为5组,经大鼠尾静脉注射不同浓度和体积的USPIO纳米颗粒,依次为1组(5 mg/ml、5 ml/kg)、2组(10 mg/ml、0.5 ml/kg)、3组(10 mg/ml、5 ml/kg)、4组(10 mg/ml、10 ml/kg)、5组(1 mg/ml、5 ml/kg)。给药前后行MR扫描,计算肿瘤组织强化率、正常组织与肿瘤组织的对比噪声比(CNR),观察1组1、5、15、30、45、60 min的肿瘤组织强化特征,分别比较不同体积组(2、3、4组)和不同浓度组(1、3、5组)的肿瘤组织强化效果。结果1组大鼠注射USPIO纳米颗粒后,肿瘤组织强化率持续升高,在45 min达到峰值,30~60 min强化率差异无统计学意义(P>0.05);CNR持续升高,在30 min达到峰值,注射前与注射后的每个时间点CNR差异均有统计学意义(P<0.05)。不同体积组中,5 ml/kg组的强化率优于0.5 ml/kg组和10 ml/kg组(P<0.05),5 ml/kg组的CNR优于0.5 ml/kg组(P<0.05),5 ml/kg组和10 ml/kg组的CNR差异无统计学意义(P>0.05)。不同浓度组中,5 mg/ml组和10 mg/ml组的强化率、CNR均优于1 mg/ml组(P<0.05)。结论大鼠注射体积为5 ml/kg、浓度为5 mg/ml的USPIO纳米颗粒后30 min进行T1WI增强扫描,肿瘤可以达到满意的强化效果。Purpose To study the changes of MRI enhancement effect with time,injection volume and concentration after injecting a new type of ultrasmall superparamagnetic iron oxide(USPIO)nanoparticles into rat glioma model,and to explore the appropriate enhancement scanning injection scheme.Materials and Methods Twenty-five rat glioma models were randomly divided into five groups.USPIO nanoparticles with different concentrations and volumes were injected into the tail vein of rats,and set in proper order for group 1(5 mg/ml,5 ml/kg),group 2(10 mg/ml,0.5 ml/kg),group 3(10 mg/ml,5 ml/kg),group 4(10 mg/ml,10 ml/kg),group 5(1 mg/ml,5 ml/kg).MRI scanning was performed before and after administration to calculate the T1 enhancement rate of tumor tissue and the contrast noise ratio(CNR)between normal tissue and tumor tissue.MR enhancement features of tumor tissue in group 1 were observed at different time points(1,5,15,30,45 and 60 minutes).MR enhancement effect was observed in different volume groups(group 2,3 and 4)and different concentration groups(group 1,3 and 5).Results After injection of USPIO nanoparticles in group 1,the T1 enhancement rate of tumor tissue increased continuously and reached the peak at 45 minutes.There was no significant difference in 30-60 minutes enhancement rate(P>0.05).CNR increased continuously and reached the peak at 30 minutes.There was significant difference in CNR at each time point before and after injection(P<0.05).In different volume groups,the enhancement rate of 5 ml/kg group was better than that of the other two groups(P<0.05).CNR of 5 ml/kg group was better than that of 0.5 ml/kg group(P<0.05),and there was no significant difference between 5 ml/kg and 10 ml/kg groups(P>0.05).In different concentration groups,the enhancement rate and CNR of 5 mg/ml and 10 mg/ml groups were better than those of 1 mg/ml group(P<0.05).Conclusion The tumor could achieve satisfactory enhancement effect when T1WI enhanced scanning is performed 30 minutes after the injection of USPIO nanoparticles with a volume of
