摘 要:目的:探讨黄芪中黄芪甲苷含量测定方法可行性。方法:采用中性氧化铝柱层析方法与药典法对黄芪药材进行处理,对两种方法所测不同批次黄芪中黄芪甲苷含量数据进行比较。结果:采用中性氧化铝柱层析方法处理,黄芪甲苷在0.2343~1.1715 mg/mL呈良好的线性关系,平均回收率为100.56%(RSD=3.28%),中性氧化铝柱层析法、《中国药典》法测定黄芪药材中黄芪甲苷含量分别为0.0740%,0.1005%,0.1302%,0.1113%,0.1520%,0.0998%,0.0793%,0.1303%和0.0694%,0.0860%,0.0854%,0.0757%,0.1054%,0.0698%,0.0402%,0.0720%。结论:采用中性氧化铝柱层析方法对黄芪药材进行处理,较药典法操作起来更简便、重复性更好。该方法可以为快速、准确测定黄芪甲苷含量提供参考。Objective:To study the feasibility of determination of astragaloside Ⅳ in Astragali Radix.Method:Neutral alumina column chromatography and Chinese Pharmacopoeia were used to treat Astragalus membranaceus,and the data of astragaloside Ⅳ in different batches of Astragalus membranaceus determined by the two methods were compared.Results:Treatment with neutral alumina column chromatography,Astragaloside Ⅳ showed a good linear relationship in the range of 0.2343~1.1715 mg/mL.The average recovery rate of Astragaloside Ⅳ was 100.56%(RSD=3.28%).The contents of astragaloside Ⅳ in Astragali Radix determined by Neutral alumina column chromatography and pharmacopoeia method were 0.0740%,0.1005%,0.1302%,0.1113%,0.1520%,0.0998%,0.0793%,0.1303%;0.0694%,0.0860%,0.0854%,0.0757%,0.1054%,0.0698%,0.0402%,0.0720%.Conclusion:Treatment of Astragalus membranaceus by neutral alumina column chromatography is simpler and more reproducible than pharmacopeia.This method can provide a reference for rapid and accurate determination of astragaloside IV.
