摘  要:目的:观察α-倒捻子素(MAN)对阿尔茨海默病(AD)大鼠认知记忆功能的作用,分析可能的作用机制。方法:采用D-半乳糖和Aβ_(1-42)双因素制备AD大鼠模型,用MAN灌胃21 d后采用Morris水迷宫实验观察大鼠的学习记忆能力,并检测血清中IL-1β、TNF-α水平,海马α7nAChR表达情况及CA1区的病理学变化。结果:MAN组大鼠的学习记忆能力较模型组获得明显改善,血清IL-1β、TNF-α的水平降低,海马α7nAChR表达显著上调并且海马CA1区神经细胞损伤较少,细胞结构基本完整。结论:MAN能够改善AD大鼠的认知记忆功能可能与其减轻体内炎症反应相关。Objective:To observe the effect ofα-mangostin(MAN)on cognitive and memory function of Alzheimer’s disease(AD)rats and analyze the possible mechanism.Methods:AD rats model was established by using D-galactose and Aβ_(1-42) in rats.Morris water maze test was used to evaluate spatial learning and memory ability,and serum IL-1β and TNF-α levels were tested.α7n ACh R expression in hippocampus and pathological changes in hippocampus CA1 region of rats was observed after 21 days of intragastric administration.Results:Compared with the model group,the cognitive and memory ability of the MAN group were significantly improved,the levels of serum IL-1β and TNF-α were decreased,the expression of α7NACh R in the hippocampus was significantly up-regulated,and the nerve cells in the hippocampus CA1 region were less damaged and the cell structure was basically intact.Conclusion:MAN can improve the cognitive and memory ability of AD model rats possibly by reducing inflammatory response and protecting nerve cells from damage.
