摘 要:【目的】研究外源海藻糖对兰州百合鳞片扦插过程碳水化合物代谢的影响,为调控鳞片繁殖技术提供理论参考。【方法】用外源海藻糖(0、10、50、100、200、500 mmol/L)处理兰州百合鳞片并进行扦插繁殖。【结果】不同浓度外源海藻糖均显著促进了百合鳞片子鳞茎发生,其中50 mmol/L的作用效果最佳。外源海藻糖处理鳞片的淀粉、蔗糖、海藻糖和可溶性糖含量显著高于对照,并在75 d时差异达到最大,分别是对照的1.6、1.8、1.4和1.9倍。说明外源海藻糖增加了百合鳞片的内源糖含量。相比于对照,外源海藻糖处理显著增加了淀粉合成相关酶ADP-葡萄糖焦磷酸化酶的活性,而蔗糖降解相关酶蔗糖合成酶的活性被明显抑制。ADP-葡萄糖焦磷酸化酶和蔗糖合成酶相关基因表达的结果和酶活的结果保持一致。此外,外源海藻糖增加了鳞茎内源海藻糖的含量,提高了其合成相关酶活性和基因表达水平。【结论】外源海藻糖调节了百合鳞片糖代谢相关酶的活性,通过增加百合鳞片糖合成和减缓糖降解促进其子鳞茎的发生。【Objective】To study the effect of exogenous trehalose on Lanzhou lily scale cutting and its carbohydrate metabolism,and to laid a foundation for regulating scale propagation technology and further studying the carbohydrate metabolism in cutting propagation.【Method】The scales of Lanzhou lily were treated with exogenous trehalose(0,10,50,100,200 and 500 mmol/L)and cutting propagation was carried out.【Result】50 mmol/L exogenous trehalose had the best effect on the production of bulblets.The content of starch,sucrose,trehalose and soluble sugar in the scales under the treatment of exogenous trehalose was significantly higher than that of the control,and the difference reached the maximum at 75 days,which were 1.6,1.8,1.4,and 1.9 times of the control,respectively.Exogenous trehalose increased the endogenous sugar content of lily scales.Compared with the control,the activity of ADPG pyrophosphorylase,an enzyme related to starch synthesis,increased significantly under trehalose treatment,while the activity of sucrose synthase,an enzyme related to sucrose degradation,was significantly inhibited.So,exogenous trehalose might affect the content of carbohydrates in scales by regulating the activity of enzymes related to sugar metabolism.The results of ADPG pyrophosphorylase and sucrose synthase at the transcription level were consistent with the results of enzyme activity.In addition,exogenous trehalose enhanced the content of endogenous trehalose,and improved its synthesis-related enzyme activity and gene expression level.【Conclusion】It can be seen that exogenous trehalose might increase the sugar content of scales by regulating the activity of enzymes related to sugar metabolism.Exogenous trehalose could promote the occurrence of lily bulbs by increasing the endogenous sugar synthesis and slowing down sugar degradation in lily scales.
