摘  要:目的探讨地氟烷对老年大鼠海马β-淀粉样蛋白(Aβ)、Tau蛋白表达及学习记忆能力的影响。方法老年雄性SD大鼠25只,采用随机数字表法,将SD大鼠分为对照(C)组与实验组,实验组分为4个亚组,每组5只。C组吸入1 L/min空气+1 L/min O_(2)30 min;实验组吸入8%地氟烷30 min,麻醉苏醒后2 h(D-2)组、12 h(D-12)组、24 h(D-24)组、72 h(D-72组)通过Morris水迷宫实验评价大鼠的空间学习记忆能力,采用免疫荧光检测大鼠海马内Aβ、Tau蛋白的表达量。结果与第1、2天比较,第3、4、5天逃避潜伏期明显缩短(P<0.05)。各组大鼠有效区域运动时间、有效区域运动距离、有效区域进入次数差异无统计学意义(P>0.05)。各组大鼠海马内Tau、Aβ蛋白的表达水平无明显差异(P>0.05)。结论地氟烷短时间内低浓度麻醉不损害老年大鼠的短期学习记忆能力。Objective To investigate the effects of desflurane on expression of Aβand Tau protein and learning and memoryability in hippocampus of aged rats.Methods Twenty-five aged male SD rats,SD rats were divided into control(C)group and experimental group by using the random number table method,The experimental group was divided into 4 subgroups with 5 rats in each group.C group inhaled 1 L/min air+1 L/min O_(2) for 30 min;Experimental group inhaled 8%desflurane for 30 min.After anesthesia recovery 2 h(D-2)group,12 h(D-12)group,24 h(D-24)group,72 h(D-72)group.The spatial learning and memory ability of rats was evaluated by Morris water maze test.The expression of Aβand Tau protein in hippocampus was detected by immunofluorescence.Results Compared with the 1st day and 2rd day,the escape latency was significantly shortened on 3rd,4th and 5th day(P<0.05).There were no statistically significant differences in the effective region movement time,effective region movement distance,effective region entry times of rats in each group(P>0.05).There were no statistically significant differences in the expression levels of Tau and Aβproteins in the hippocampus of rats in each group(P>0.05).Conclusions Low concentration anesthesia of desflurane in short time could not impair the short-term learning and memory ability of the aged rats.
