摘 要:为了探究谷子脂肪氧化酶(LOX)与小米储藏期间褪色特性的关系,为培育耐储藏的谷子品种提供理论依据,通过对比4种名优谷子品种豫谷1号(YG1)、龙谷25号(LG25)、沁黄2号(QH2)和晋谷21号(JG21)储藏期米色、总类胡萝卜素含量、LOX酶活性及LOX基因的表达,初步探索这4个谷子品种在储藏期间的褪色特性。结果表明,储藏初期米色b*值最大的是LG25,最小的是YG1;到储藏末期,褪色较快的谷子品种是YG1,褪色较慢的谷子品种是JG21;总类胡萝卜素降解较快的谷子品种是YG1,较慢的谷子品种是JG21;谷子品种LG25、QH2和YG1中LOX活性均呈现出先增高后降低的趋势,而谷子品种JG21的LOX活性变化程度较小。qRTPCR结果显示,在灌浆末期,SiLOX3、SiLOX4、SiLOX7基因在谷子品种YG1中的表达量最高,推测SiLOX基因的高表达是造成小米总类胡萝卜素降解较快以及褪色快的主要原因。综上所述,谷子中LOX在小米储藏褪色过程中发挥着重要作用。To explore the relationship between lipoxygenase(Lipoxygenase,LOX)and the discoloration characteristics of foxtail millet during storage,provide theoretical foundation for breeding for foxtail millet varieties with strong storage tolerance,in this study,by comparison of millet color,the total carotenoid contents,the LOX activity,and the expression profiles of LOX genes of four famous millet varieties of Yugu 1(YG1),Longgu 25(LG25),Qinhuang 2(QH2),and Jingu 21(JG21)during storage,discoloration characteristics of the four varieties during storage were preliminarily explored.The results showed that the b*values of millet color of LG25 was the highest and the value of YG1 was the lowest at the early storage.At the end of storage,the millet variety with the fastest discoloration was YG1,while the slowest one was JG21.The variety with the faster degradation of the total carotenoid was YG1,and the slower one was JG21.The activity of LOX in LG25,QH2,and YG1 increased at first then decreased,but the activity of LOX in JG21 changed slightly.The results of qRT-PCR showed that the expression of SiLOX3,SiLOX4,SiLOX7 genes in YG1 was the highest at late filling stage.It was speculated that the high expression of SiLOX gene was the main reason for the rapid degradation of total carotenoid and rapid discoloration of millet.To sum up,LOX in millet plays an important role in the discoloration process of millet storage.
