摘 要:目的研究异土木香内酯对宫颈癌细胞的迁移、侵袭的影响及其机制。方法体外培养人宫颈癌HeLa细胞株,随机分为对照组、低剂量实验组、中剂量实验组、高剂量实验组和AG490组。低、中、高剂量实验组分别用10,40和80μmol·L^(-1)异土木香内酯处理细胞,AG490组用25μmol·L^(-1)的AG490处理细胞,对照组加入等体积二甲基亚砜。通过划痕实验检测细胞的迁移能力;用侵袭实验(Transwell)检测细胞的侵袭能力;用蛋白质印迹(Western blot)法检测蛋白表达。结果对照组和低、中、高剂量实验组细胞的迁移率分别为(47.62±4.81)%,(42.53±4.22)%,(36.88±3.70)%和(12.45±1.25)%;细胞侵袭数目分别为(98.34±9.85),(92.33±9.24),(71.64±7.22)和(36.51±3.62)个;N-cadherin蛋白的相对表达水平分别为1.05±0.11,0.97±0.09,0.77±0.08和0.59±0.06;Vimentin蛋白的相对表达水平分别为1.03±0.10,0.92±0.09,0.72±0.07和0.53±0.04;E-cadherin蛋白的相对表达水平分别为0.23±0.03,0.31±0.04,1.37±0.14和1.69±0.17;p-JAK2蛋白的相对表达水平分别为1.02±0.10,0.96±0.10,0.73±0.07和0.54±0.06;JAK2蛋白的相对表达水平分别为1.03±0.10,0.98±0.10,0.96±0.10和0.95±0.09;p-STAT3蛋白的相对表达水平分别为1.02±0.10,0.97±0.10,0.75±0.08和0.46±0.05;STAT3蛋白的相对表达水平分别为1.03±0.10,0.96±0.10,0.94±0.09和0.95±0.10。以上指标,中、高剂量实验组与对照组比较,差异均有统计学意义(均P<0.05)。结论异土木香内酯可通过下调p-JAK2和p-STAT3蛋白表达抑制HeLa细胞的迁移、侵袭及上皮间质转化。Objective To investigate the effect of isoalantolactone on the migration and invasion of cervical cancer cells and its mechanism.Methods Human cervical cancer cell lines HeLa were cultured in vitro and randomly divided into control group,experimental-L group,experimental-M group,experimental-H group and AG490 group.The cells in experimental-L,experimental-M and experimental-H groups were treated with 10,40 and 80μmol·L^(-1) isoalantolactone,respectively.The cells in AG490 group were treated with 25μmol·L^(-1) AG490.The migration ability of cells was detected by wound healing assay.Transwell was used to detect the invasion of cells.Western blot assay was used to detect the expression of proteins.Results The cell migration rates of control group,experimental-L group,experimental-M group and experimental-H group were(47.62±4.81)%,(42.53±4.22)%,(36.88±3.70)%and(12.45±1.25)%,the number of invaded cells were 98.34±9.85,92.33±9.24,71.64±7.22 and 36.51±3.62,the relative expression levels of N-cadherin protein were 1.05±0.11,0.97±0.09,0.77±0.08 and 0.59±0.06,the relative expression levels of Vimentin were 1.03±0.10,0.92±0.09,0.72±0.07 and 0.53±0.04,the relative expression levels of E-cadherin protein were 0.23±0.03,0.31±0.04,1.37±0.14 and 1.69±0.17,the relative expression levels of phosphorylated Janus kinase 2(p-JAK2)protein were 1.02±0.10,0.96±0.10,0.73±0.07 and 0.54±0.06,the relative expression levels of JAK2 protein were 1.03±0.10,0.98±0.10,0.96±0.10 and 0.95±0.09,the relative expression levels of phosphorylated signal transducer and transcriptional activator 3(p-STAT3)protein were 1.02±0.10,0.97±0.10,0.75±0.08 and 0.46±0.05,the relative expression levels of STAT3 protein were 1.03±0.10,0.96±0.10,0.94±0.09 and 0.95±0.10.There was significant difference between experimental-M group,experimental-H group and control group(all P<0.05).Conclusion Isoalantolactone can inhibit the migration,invasion and epithelial mesenchymal transformation of HeLa cells by down-regulating the expressi
