摘 要:多肽药物有特异性好、生物活性高和不良反应少等优点。随着多肽合成技术的日益完善,为了解决多肽口服生物利用度较低的问题,多肽口服给药技术成为热点研究方向。多肽药物口服给药系统的设计有三个关键点:提高多肽抗酶解能力、帮助多肽通过黏膜屏障以及促进多肽药物吸收。目前不少多肽药物口服制剂已在临床试验阶段并获得上市许可,在口服多肽药物开发中许多技术展现了良好的临床应用前景。文章介绍了目前口服多肽药物应用技术的机制和特点,综述近年来发展的新型口服多肽临床应用。Peptide drugs have the advantages of high specificity,high biological activity and few adverse reactions.With the increa-sing improvement of peptide synthesis technology,in order to solve the problem of low oral bioavailability of peptide drugs,oral peptide administration technology has become a hot research direction.There are three key points in the design of the oral peptide delivery system:improving the ability of the resistance of polypeptide to enzymatic hydrolysis,assisting peptides pass through the mucosal barrier and promoting peptides absorption.At present,many oral peptide drugs are in clinical trials,and some oral peptide drugs have been approved for marketing.Many techniques have shown good clinical application prospects in the development of oral polypeptide drugs.The mechanism and characteristics of the technology used in oral peptide drugs were introduced,and the oral clinical applications of new peptides developed was reviewed in recent years.
