摘  要:目的:优化五味子藤茎中木脂素的提取纯化工艺。方法:在正交试验设计的基础上,以乙醇体积分数、提取时间、溶剂倍数、提取次数为影响因素,考察五味子藤茎中木脂素的最佳提取条件。利用单因素考察法,以最佳树脂类型、上样液浓度、最大上样量、洗脱剂(乙醇)体积分数、洗脱剂用量为影响因素,考察最佳的木脂素纯化条件。结果:最佳提取工艺为8倍量95%乙醇提取2次,每次提取1h。最佳纯化工艺为AB-8型大孔吸附树脂,上样液木脂素浓度2.0mg/mL,最大上样量70mL,洗脱剂体积分数95%,洗脱剂用量为80mL。结论:该方法操作简单,结果稳定,为进一步研发新药奠定基础。Objective:To optimize the extraction and purification of lignans from rattan stems of Schisandra chinensis.Methods:Based on the orthogonal test design,the optimal extraction conditions of lignans from ratten stems of Schisandra chinensis were investigated by the factors of ethanol volume fraction,extraction time,solvent multiple and extraction times.The optimal purification conditions of lignans were investigated by single factor method,with the best resin type,concentration of sample solution,maximum sample loading,volume fraction of eluent(ethanol)and eluent dosage as the influencing factors.Results:The optimal extraction process was 8 times the amount of 95%ethanol,twice extraction,each time 1 hour.The best purification process was with AB-8 macroporous resin,the concentration of lignans in the loading solution was 2.0 mg/mL,the maximum loading capacity was 70 mL,the volume fraction of eluent was 95%,and the amount of eluent was 80 mL.Conclusion:This method is simple to operate and has stable results,which is a significant basis for further development of new drugs.
