摘  要:目的探究低强度超声(LIUS)联合骨髓间充质干细胞(BMSCs)对大鼠皮肤创伤增殖期的作用及机制。方法在大鼠背部剪出直径为2cm的圆形伤口建立全层皮肤切除模型,设置LIUS组(0.2W/cm^(2),20min/d,7d)、BMSCs组、LIUS+BMSCs组和对照组。观察大鼠皮肤创伤愈合情况并通过Image J计算皮肤创伤愈合率;HE染色观察3d和7d大鼠皮肤创面组织病理变化情况;ELISA检测大鼠血清0、3、7、14和28d基质细胞衍生因子(SDF-1)、肿瘤坏死因子(TNF-α)、血小板衍生生长因子(PDGF)的含量。结果与其他组相比,LIUS+BMSCs组能够提高大鼠皮肤创伤愈合速度;3d时可见肉芽组织开始形成,7d时LIUS+BMSCs组肉芽组织中新生血管明显,有统计学意义(P<0.05);大鼠血清中TNF-α和PDGF含量呈先上升后下降趋势,SDF-1含量呈下降趋势,与其他组相比,LIUS+BMSCs组在第3天和第7天都存在统计学意义(P<0.05)。结论LIUS联合BMSCs能够缩短大鼠皮肤愈合增殖期,其机制可能是影响了皮肤愈合有关的细胞因子分泌和合成。Objective To investigate the effect and mechanism of low intensity ultrasound(LIUS)combined with bone marrow mesenchymal stem cells(BMSCs)on the proliferative stage of skin trauma in rats.Methods The model of full-thickness skin resection was established by cutting a circular wound with a diameter of 2 cm on the back of rats.The LIUS group(0.2 W/cm^(2),20 min,7 d),BMSCs group,LIUS+BMSCs group and control group were set up.The skin wound healing of rats was observed,and the skin wound healing rate was calculated by Image J.HE staining was used to observe the histopathological changes of skin wounds in rats at 3 days and 7 days old.The levels of stromal cell derived factor(SDF-1),tumor necrosis factor(TNF-α)and platelet-derived growth factor(PDGF)in serum of rats at 0,3,7,14 and 28 days were determined by ELISA.Results Compared with other groups,LIUS+BMSCs group can improve the rate of skin wound healing in rats;Granulation tissue began to form at day 3.And on day 7,angiogenesis was significant in the LIUS+BMSCs group(P<0.05).The contents of TNF-αand PDGF in serum of rats increased first and then decreased,while the content of SDF-1 decreased.Compared with other groups,there were statistical differences in LIUS+BMSCs group on days 3 and 7(P<0.05).Conclusions LIUS combined with BMSCs can shorten the proliferation period of skin healing in rats,possibly achieve by affecting the secretion and synthesis of cytokines in the healing process.
