摘  要:为了解猕猴桃铁超氧化物歧化酶基因的序列特征及其在果实采后的表达特性,采用RT-PCR进行基因克隆并利用qRT-PCR分析其在不同贮藏温度和激素处理后的表达特征。结果显示,AdFSD1的开放阅读框为915 bp,编码304个氨基酸,GenBank登录号为KY471360,其基因组包含9个外显子和8个内含子,在进化树中与茶树Fe-SOD的亲缘关系最近。定量结果表明,AdFSD1在根部组织的表达量最大。果实中AdFSD1基因在低温贮藏后下调表达。采后脱落酸处理诱导AdFSD1的表达,但赤霉素处理抑制AdFSD1的表达。研究结果为后续探究超氧化物歧化酶与猕猴桃果实采后品质的关系奠定基础。To investigate the sequence characteristics of the iron superoxide dismutase from Actinidia chinensis and its postharvest expression patterns,RT-PCR was used for gene cloning and qRT-PCR was used to analyze its expression patterns after storage at different temperatures and treatments with hormones.The results showed that the open reading frame of AdFSD1 was 915 bp,encoding 304 amino acids,and the GenBank accession number was KY471360.Its genome contained 9 exons and 8 introns.AdFSD1 had the closest relationship with Fe-SOD of Camellia sinensis in the phylogenetic tree.Quantitative results showed that AdFSD1 had the largest expression level in root tissues.The expression of AdFSD1 gene in fruits was down-regulated after storage at low temperature.Postharvest treatment with abscisic acid induced the expression of AdFSD1,but treatment with gibberellin inhibited the expression of Ad FSD1.These results laid the foundation for the subsequent exploration of the relationship between superoxide dismutase and the postharvest quality of kiwifruit.
