摘  要:目的:观察丰富环境干预对缺血再灌注脑损伤大鼠的血脑屏障结构及其功能的影响。方法:采用线栓栓塞大鼠大脑中动脉的方法制备缺血再灌注脑损伤模型,经丰富环境干预2周,采用神经功能7分评分方法对缺血再灌注脑损伤大鼠进行神经功能的缺失评分,运用透射电镜拍照观察大鼠梗死皮层周边区血脑屏障的超微结构变化情况,采用股静脉注射Evans蓝染液外渗法检测血脑屏障通透性的变化情况,以及免疫荧光染色法观察星形胶质细胞表达情况。结果:Evans蓝染液结果显示丰富环境干预可明显修复血脑屏障的通透性,表现为降低Evans蓝染液外渗至大鼠大脑实质中的量,电镜结果显示丰富环境干预可显著改善梗死皮层周边区血脑屏障结构的完整性,并且显著减轻了皮层血管周边的星形胶质细胞和周细胞的损伤和水肿程度,且丰富环境干预可促进皮层星形胶质细胞的表达,保持血脑屏障结构和功能的完整性。结论:丰富环境干预可修复缺血再灌注脑损伤大鼠引发的血脑屏障结构和功能的破坏,从而促进神经功能的恢复,其可能与星形胶质细胞的表达有关。Objective:To observe the effects of enriched environment on the structure and permeability of blood-brain barrier in rats with ischemia-reperfusion injury.Method:The model of ischemia-reperfusion brain injury was prepared by tMCAO method.After enriched environment intervention for 2 weeks,the rats were scored for neurological deficit by 7-point scoring method.The ultrastructural changes of blood-brain barrier were observed by transmission electron microscope,the permeability changes of blood-brain barrier were observed by intravenous Evans blue extravasation,and the expression of astrocytes was observed by immunofluorescence.Result:Enriched environment significantly improved the permeability of blood-brain barrier,which was manifested by reduced distribution of Evans blue in the brain.The ultrastructure of blood-brain barrier showed that enriched environment significantly improved the integrity of blood-brain barrier,significantly reduced the swelling of perivascular astrocytes and pericytes.In addition,enriched environment promoted the expression of astrocytes and maintained the integrity of the structure and function of blood-brain barrier.Conclusion:Enriched environment could repair the damage of blood-brain barrier structure and function caused by ischemia-reperfusion brain injury in rats,and promote the recovery of neural function,which may be related to the expression of astrocytes.
