摘 要:目的研究黄芩苷对糖尿病鼠的心肌保护机制。方法取24只雄性SD鼠随机分为对照组(6只)、黄芩苷对照组(6只)、模型组(6只)、治疗组(6只)。模型组和治疗组尾静脉注射45 mg/kg链脲佐菌素建立实验性糖尿病模型。黄芩对照组和治疗组采用腹腔注射黄芩苷药液15μmol/(L·kg·d)方法;对照组和模型组采用腹腔注射生理盐水方法,四组均用正常普通饲料饲养。8周后比较四组的血糖、线粒体结构、心肌线粒体损伤比例、呼吸链复合物Ⅰ、Ⅱ、Ⅲ和Ⅳ的活性、ND1 mRNA和ND1蛋白表达。结果模型组的血糖高于对照组,治疗组的血糖低于模型组,差异有统计学意义(P<0.05)。模型组的损伤线粒体百分比高于对照组,治疗组的损伤线粒体百分比低于模型组,差异有统计学意义(P<0.05)。模型组的心肌线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性低于对照组,治疗组的心肌线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性高于模型组,差异有统计学意义(P<0.05)。模型组的ND1 mRNA和ND1蛋白低于对照组,治疗组的ND1 mRNA和ND1蛋白高于模型组,差异有统计学意义(P<0.05)。结论糖尿病鼠的心肌线粒体呼吸链复合物的活性及粒体呼吸链复合物Ⅰ亚基ND1基因和蛋白表达均降低,黄芩苷可能通过提高线粒体呼吸链复合物活性和增强复合物Ⅰ亚基ND1基因和蛋白表达参与对糖尿病鼠心肌的保护作用。Objective To study the protective mechanism of Baicalin on the myocardium of diabetic rats.Methods Twenty-four male SD rats were randomly divided into the control group(6 rats),the Baicalin control group(6 rats),the model group(6 rats)and the treatment group(6 rats).Experimental diabetes models were established by tail vein injection of 45 mg/kg streptozotocin in the model group and the treatment group.The Baicalin control group and the treatment group were given intraperitoneal injection of 15μmol/(L·kg·d)Baicalin liquid,while in the control group and the model group,the intraperitoneal injection of normal saline was used.The four groups were fed with normal diet.After 8 weeks,blood glucose,mitochondrial structure,myocardial mitochondrial damage ratio,activity of respiratory chain complexesⅠ,Ⅱ,Ⅲ,Ⅳ,ND1 mRNA and ND1 protein expression were compared among the four groups.Results The blood glucose of the model group was higher than that of the control group,and the blood glucose of the treatment group was lower than that of the model group,the differences were statistically significant(P<0.05).The percentage of damaged mitochondria in the model group was higher than that in the control group,and the percentage of damaged mitochondria in the treatment group was lower than that in the model group,and the differences were statistically significant(P<0.05).The activities of myocardial mitochondrial respiratory chain complexesⅠ,Ⅱ,Ⅲ,Ⅳin the model group were lower than those in the control group,the activities of myocardial mitochondrial respiratory chain complexesⅠ,Ⅱ,Ⅲ,Ⅳin the treatment group were higher than those in the model group,the differences were statistically significant(P<0.05).The ND1 mRNA and ND1 protein in the model group were lower than those in the control group,and the ND1 mRNA and ND1 protein in the treatment group were higher than those in the model group,the differences were statistically significant(P<0.05).Conclusion The activity of mitochondrial respiratory chain complex an
