摘  要:目的:观察电针对克罗恩病(CD)大鼠结肠组织AIM2炎症通路和自噬相关分子的调节作用。方法:将SD大鼠随机分为正常组、模型组、电针组、西药组,每组8只。采用TNBS诱导CD大鼠模型,电针组予电针气海穴和双侧天枢穴,西药组予柳氮磺吡啶肠溶片灌胃。检测结肠组织AIM2、p62、FliⅠ、Nrf2、LC3Ⅱ、ASC、Caspase-1、IL-18蛋白和/或mRNA表达,以及血清ICAM-1、VCAM-1和E-Selectin的浓度。结果:模型组结肠HE染色可见溃疡,电针组、西药组结肠上皮覆盖较完整,可见愈合性溃疡。与模型组比较,电针组、西药组血清ICAM-1、VCAM-1、E-Selectin浓度均显著降低(P<0.01),结肠组织p62、FliⅠ、Nrf2蛋白表达显著升高(P<0.01),AIM2、LC3Ⅱ、Caspase-1、IL-18蛋白表达显著降低(P<0.01),电针组、西药组结肠组织LC3Ⅱ、AIM2、ASC mRNA表达均显著降低(P<0.05,P<0.01),西药组结肠组织p62 mRNA表达显著升高(P<0.01)。结论:电针能调节结肠组织自噬相关分子表达,抑制AIM2炎症通路,并能降低血清黏附分子浓度,可能是电针减轻克罗恩病结肠炎症反应的效应机制之一。Objective:To investigate the regulatory effects of electroacupuncture on the inflammatory pathway of AIM2 and autophagy regulation in the colon tissue of Crohn’s disease(CD)rats.Methods:SD rats were randomly divided into normal group,model group,electroacupuncture group and western medicine group,and the electroacupuncture group was given electroacupuncture points at Qihai(RN 6)and Tianshu(ST 25)on both sides,while the western medicine group was given salazosulfapyridine intestinal tablets by gavage.The protein and/or mRNA expression of AIM2,p62,FliI,Nrf2,LC3II,Caspase-1,IL-18 in the colon tissue,and the concentration of serum ICAM-1,VCAM-1 and E-Selectin were detected.Results:Ulcers were visible in the HE-stained colon of the model group,while healing ulcers were visible in the electroacupuncture and western medicine groups with more complete colonic epithelial coverage.Compared with the model group,serum concentrations of ICAM-1,VCAM-1 and E-Selectin were decreased in the electroacupuncture group and western medicine group(P<0.01).The protein expression levels of p62,FliI and Nrf2 in colon tissue were significantly increased(P<0.01),while the protein expression levels of AIM2,LC3Ⅱ,Caspase-1 and IL-18 were significantly decreased(P<0.01).The mRNA expression levels of AIM2,LC3II and ASC in colon tissue was decreased in the electroacupuncture and western medicine groups(P<0.05,P<0.01),and the mRNA expression level of p62 in colon tissue was significantly increased in the western medicine group(P<0.01).Conclusion:Electroacupuncture can regulate the expression of autophagy-related proteins and AIM2 inflammatory pathway in colon tissue,and reduce the concentrations of serum adhesion molecules,which may be the underlying mechanisms of electroacupuncture to reduce the inflammatory response of colon in CD.
