摘  要:目的:探究小儿推拿退热六法对脂多糖(LPS)致热幼兔的退热疗效及退热机制。方法:将30只新西兰幼兔随机分为盐水组、模型组和推拿组。模型组和推拿组通过耳缘静脉注射LPS建立发热模型。推拿组在造模后1 h行退热六法(开天门、推坎宫、揉太阳、揉耳后高骨、清天河水、推脊)干预,实验过程中监测肛温。造模后3 h,取下丘脑进行液相色谱-质谱联用(LC-MS)非靶向代谢组学研究。结果:造模后2 h和3 h,推拿组肛温显著低于模型组(P<0.05)。代谢组学结果显示盐水组与模型组共有171个差异代谢物,富集到123条通路上;推拿组与模型组共有116个差异代谢物,富集到mTOR、PI3K-AKT、GABA能突触等77条通路上。结论:退热六法可降低发热幼兔体温,其退热机制可能与调节糖代谢、脂质代谢有关。Objective:To explore the therapeutic effect and antipyretic mechanism of six antipyretic manipulations of pedeatric tuina on lipopolysaccharide(LPS)-induced fever in young rabbits.Methods:Thirty New Zealand young rabbits were randomly divided into:saline group,model group and tuina group.The fever model was established by injecting LPS into ear vein in model group and tuina group.The tuina group was intervened with the‘six antipyretic manipulations’(pushing Tianmen,pushing Kangong,kneading Taiyang,kneading Erhougaogu,clearing Tianheshui and pushing the Spine)1 h after modeling.The anal temperature was monitored in real time during the experiment.3 h after modeling,hypothalamus was taken for non-targeted metabolomics study by liquid chromatography-mass spectrometry(LC-MS).Results:Two and three hous after modeling,the anal temperature in tuina group was significantly lower than that in model group(P<0.05).Metabolomics results showed that there were 171 differential metabolites in saline group and model group,which were enriched in 123 pathways.There were 116 differential metabolites in tuina group and model group,which were enriched in 77 pathways,such as mTOR pathway,PI3K-AKT pathway,and GABAergic synapse and so on.Conclusion:The six antipyretic manipulations can reduce the body temperature of febrile rabbits,and its antipyretic mechanism may be related to the regulation of carbohydrate metabolism and lipid metabolism.
