摘  要:目的:研究电针预处理血海、曲池穴对荨麻疹大鼠模型PI3K/PDK1/AKT通路相关蛋白表达的影响。方法:32只健康SD大鼠随机分为空白组、模型组、电针组、西药组,每组8只。电针组采用电针双侧血海、曲池穴,电流1 mA、疏密波、频率2 Hz,留针20 min,1次/d;西药组采用氯雷他定稀释液(1 mg/kg)灌胃,1次/d;空白组、模型组给予同等剂量的0.9%氯化钠溶液(1 mg/kg)灌胃,1次/d。另取3只健康SD大鼠,采用鸡卵白蛋白与免疫佐剂(卡介苗)联合注射法制备同种异体大鼠抗卵白蛋白血清。除空白组外,其余组均建立荨麻疹大鼠模型。直尺测量大鼠皮肤蓝斑直径,甲苯胺蓝染色法检测皮肤肥大细胞脱颗粒形态及数目,HE染色观察皮肤病理变化,ELISA法检测血清白细胞介素-4(IL-4)表达,免疫组织化学法检测皮下组织IgE、组胺以及皮肤组织磷脂酰肌醇3-激酶(PI3K)、磷酸肌醇依赖性激酶1(PDK1)、蛋白激酶B(AKT)表达水平。结果:与空白组比较,模型组大鼠皮肤蓝斑直径、皮肤组织炎症细胞浸润、肥大细胞脱颗粒率、皮下组织IgE和组胺,以及皮肤肥大细胞PI3K、PDK1、AKT和血清IL-4显著增多(P<0.01);与模型组比较,电针组、西药组大鼠以上指标显著减少(P<0.05,P<0.01)。结论:电针预处理血海穴、曲池穴可减小荨麻疹动物模型蓝斑直径,改善皮肤炎症反应,减少肥大细胞脱颗粒,且该效应与调控IgE介导的PI3K/PDK1/AKT信号通路激活和组胺、IL-4表达有关。Objective:To study the effects of electroacupuncture pretreatment at Xuehai(SP 10)and Quchi(LI 11)points on the expression of PI3K/PDK1/AKT pathway related proteins in urticaria rat model.Methods:Thirty-two healthy SD rats were randomly divided into blank group,model group,electroacupuncture group and Western medicine group,with 8 rats in each group.The acupuncture group was treated with electroacupuncture at Xuehai(SP 10)and Quchi(LI 11)on both sides.The electroacupuncture parameters were current 1 mA,density wave,frequency 2 Hz,needle retention for 20 min,once a day.In the western medicine group,loratadine diluent(1 mg/kg)was administered by gavage once a day.The blank group and the model group were given the same dose of normal saline(1 mg/kg)by gavage once a day.In addition,3 healthy SD rats were selected to prepare allogeneic rat anti ovalbumin serum by combined injection of chicken ovalbumin and immune adjuvant(BCG).Except the blank group,urticaria rat models were established in other groups.The diameter of locus coeruleus in rat skin were measured by rulers,morphology and number of mast cell degranulation were detected by toluidine blue staining,skin pathological changes were observed by HE staining,the expression levels of serum IL-4 were detected by ELISA,subcutaneous IgE,histamine and the expression levels of PI3K,PDK1 and AKT in skin were detected by immunohistochemistry.Results:Compared with the blank group,the diameter of locus coeruleus of rat,the infiltration of inflammatory cells in the model group in skin tissue,the degranulation rate of mast cells,IgE and histamine in subcutaneous tissue,PI3K,PDK1,AKT and serum IL-4 in skin mast cells in the model group increased significantly(P<0.01).Compared with the model group,the above indexes of rats in electroacupuncture group and Western medicine group decreased significantly(P<0.05,P<0.01).Conclusion:Electroacupuncture pretreatment at Xuehai(SP 10)and Quchi(LI 11)can reduce the diameter of locus coeruleus in urticaria animal model,improve skin inflammat
