摘 要:目的:探究连夏宁心方对心肌梗死(以下简称心梗)后大鼠左室重构的作用及对心房利钠肽前体蛋白A(NP PA)的影响。方法:将SD雄性大鼠随机分为空白组、假手术组、模型组和连夏宁心方组,每组6只。通过结扎大鼠冠状动脉左前降支,建立心梗模型。连夏宁心方组予连夏宁心方颗粒剂(1.4 g·kg^(-1)·d^(-1)),假手术组、模型组予0.9%氯化钠溶液,空白组不予干预。灌胃4周后,观察大鼠的一般情况、体质量变化量、心脏指数,用超声心动评估心室收缩、舒张情况,用HE染色和Masson染色观察心肌组织形态,Western blot检测心肌组织中心房利钠肽前体蛋白A(NPPA)的表达。结果:与模型组比较,连夏宁心方组大鼠一般情况得到改善,体质量增长显著增多(P<0.05),心脏指数显著减少(P<0.01);LVEF、LVFS显著增加(P<0.01),左心室收缩期指标变化显著减小(P<0.01),舒张期指标也有减小趋势;病理学结果显示连夏宁心方组较模型组大鼠心肌细胞变性较少,肌丝相对完整,间质相对均匀,胶原纤维显著减少;Western blot示连夏宁心方组NPPA表达显著减少(P<0.01)。结论:连夏宁心方的干预可抑制心梗后大鼠心肌组织的NPPA蛋白表达,减轻心梗组织炎症反应和纤维化,进而逆转左心室重构并改善左心室功能,提高心梗后的生活质量。Objective:To explore the effects of Lianxia Ningxin Formula(LXNX)on left ventricular remodeling and natriuretic peptide precursor protein A(NPPA)in rats after myocardial infarction(MI).Methods:The SD male rats were divided into control group,sham group,model group and LXNX group,6 in each group.The myocardial infarction model was established by ligating the left anterior descending coronary artery of SD rats.LXNX group was given LXNX(1.4 g·kg^(-1)·d^(-1)),sham group and model group were given 0.9%sodium chloride solution,while no intervention was given in the control group.Four weeks after gavage,the general situation,weight change and cardiac index of rats were observed.The ventricular contraction and relaxation were evaluated by echocardiography.The myocardial morphology was observed by HE staining and Masson staining.The expression of NPPA in myocardial tissue was detected by Western blot.Results:Compared with the model group,the general condition of rats in LXNX group was improved,the weight gain was significantly increased(P<0.05),and the cardiac index was significantly reduced(P<0.01);LVEF and LVFS increased significantly(P<0.01),left ventricular systolic indexes decreased significantly(P<0.01),and diastolic indexes also decreased;The pathological results showed that myocardial cells in LXNX group had less degeneration compared to the model group,muscle filaments were relatively complete,stroma was relatively uniform,and collagen fibers were significantly reduced;Western blot also showed that the expression of NPPA decreased significantly in LXNX group(P<0.01).Conclusion:The intervention of LXNX can inhibit the expression of NPPA protein in myocardial tissue of rats after AMI,reduce the inflammatory reaction and fibrosis of myocardial infarction tissue,reverse left ventricular remodeling,improve left ventricular function and improve the quality of life of rats after myocardial infarction.
