摘  要:目的:探讨魄不安于肺不寐大鼠血清生物钟分子血清褪黑素(MT)、脑褪黑素相关受体(GPR50)和松果体的变化。方法:将24只SD大鼠随机分为模型组和正常组,每组12只。采用狭长多平台浅水环境法剥夺大鼠深睡眠42 d制备慢性睡眠剥夺模型。称取大鼠体质量,进行水迷宫检测、戊巴比妥钠诱导的睡眠实验和脑肌电监测,以E L I SA检测血清M T水平,以免疫组化法检测脑和脑干G P R50的水平,透射电子显微镜观察松果体的病理变化。结果:与正常组比较,模型组大鼠体质量显著减少(P<0.05,P<0.01)、上台潜伏期显著延长(P<0.05)、穿越原平台上方次数显著减少(P<0.05);戊巴比妥钠致睡眠潜伏期显著延长(P<0.05)、睡眠时间显著缩短(P<0.05);脑电图慢波睡眠第2期及快动眼睡眠比例显著减少(P<0.01);血清MT显著降低(P<0.05);脑及脑干GPR50表达显著升高(P<0.01,P<0.05);松果体超微结构出现明显病理变化。结论:狭长多平台浅水环境法可以剥夺大鼠部分深睡眠,引起大鼠记忆、睡眠行为改变,褪黑素和GPR50及松果体超微结构的变化。上述模型指标的变化可以用于研究魄不安于肺型不寐时参考。Objective:To investigate the changes of biological clock molecule serum melatonin(MT),brain G-proteincoupled receptor 50(GPR50)and pineal gland in chronic sleep deprivation rats with corporeal soul unrest in the lung.Methods:Twenty-four SD rats were randomly divided into model group and normal group,12 in each one.The chronic sleep deprivation models were established by depriving rats of deep sleep for 42 days in the water environment with multiple narrow and long platforms in it.The body mass of rats was weighed.Water maze detection,pentobarbital sodium-induced sleep test and electroencephalogram/electromyogram(EEG/EMG)monitoring were carried out.The level of serum MT was detected by Enzyme linked immunosorbent assay(ELISA).The level of GPR50 in brain and brainstem was detected by immunohistochemistry,and the pathological changes of pineal gland were observed by transmission electron microscope.Results:Compared with the normal group,the increase of body weight decreased(P<0.05,P<0.01),the latency of getting on the platform increased(P<0.05),the times of crossing the water over the removed platform decreased(P<0.05),pentobarbital sodium-induced sleep latency extended(P<0.05),the sleep time shortened(P<0.05),the EEG/EMG proportion of slow wave sleep period 2 and REM decreased(P<0.01,P<0.05),the serum MT decreased(P<0.05),GPR50 increased(P<0.05,P<0.01),and there were significant pathological changes in the ultra microstructure of pineal gland.Conclusion:Multiple long and narrow platforms in the water environment can be used to deprive rats of part deep sleep,produce behavioral changes of memory and sleep,level changes of MT and GPR50,as well as pathological change in the ultra microstructure of pineal gland.The changes of above model rats can be used as a reference in studying the insomnia of corporeal soul unrest in the lung.
