摘　　要：目的通过观察二神丸对溃疡性结肠炎(UC)大鼠炎性反应及血小板的影响,探讨其对UC的治疗作用及相关作用机制。方法在70只SD大鼠中随机取10只作为正常对照组,其余60只采用2,4,6-三硝基苯磺酸钠(TNBS)-乙醇灌肠法建立UC模型,再将造模成功的大鼠随机分为模型组(10只)、阳性药物组(8只)、二神丸低剂量组(10只)、二神丸高剂量组(10只),其中阳性药物组予美沙拉秦缓释颗粒混悬液0.5 g/(kg·d)灌胃,二神丸低剂量组予二神丸水煎液1.575 g/(kg·d)灌胃,二神丸高剂量组予二神丸水煎液6.3 g/(kg·d)灌胃,正常对照组与模型组给予等容积蒸馏水10 mL/(kg·d)灌胃,各组均连续灌胃14 d。比较各组灌胃结束后全血血小板计数(PLT)、平均血小板体积(MPV)水平情况,血清P-选择素(P-selectin)、一氧化氮(NO)水平情况,结肠组织白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平情况。结果与正常对照组比较,模型组大鼠灌胃后全血PLT增加(P<0.05),MPV降低(P<0.05);与模型组比较,二神丸低、高剂量组大鼠全血PLT均降低(P<0.05),二神丸低剂量组MPV增加(P<0.05),阳性药物组大鼠全血PLT、MPV和二神丸高剂量组MPV与模型组比较差异均无统计学意义(P>0.05);二神丸低、高剂量组全血PLT、MPV组间比较差异均无统计学意义(P>0.05)。与正常对照组比较,模型组血清P-selectin、NO水平均增加(P<0.05);与模型组比较,阳性药物组、二神丸低剂量组、二神丸高剂量组血清NO水平和二神丸高剂量组P-selectin水平均降低(P<0.05),阳性药物组和二神丸低剂量组P-selectin水平与模型组比较差异无统计学意义(P>0.05);二神丸低、高剂量组大鼠血清P-selectin、NO水平组间比较差异均无统计学意义(P>0.05)。与正常对照组比较,模型组大鼠结肠组织IL-1β、TNF-α水平均升高(P<0.05);与模型组比较,阳性药物组、二神丸低剂量组、二神丸高剂量组大鼠结肠组织IL-1β、TNF-Objective To observe the effect of Ershen Pill on inflammatory response and platelet in ulcerative colitis(UC)rats to explore its therapeutic effect and mechanism.Methods Ten were randomly subjected from 70 SD rats as the control group(normal saline,10 mL·kg^(-1)·d^(-1)),the UV model was established by a 2,4,6-trinitrobenzene sulfonic acid(TNBS)-ethanol enema in remaining rat,the successful model rats were randomly assigned into four groups,the model group(n=10,normal saline,10 mL·kg^(-1)·d^(-1)),the positive drug group(n=8,mesalazine slow-release suspension,0.5 g·kg^(-1)·d^(-1)),and low-,and high-dose(1.575,6.3 g·kg^(-1)·d^(-1),respectively;10 cases in each)Ershen Pill groups.A 14-day continuous gavage was performed,aiming to compare platelet count(PLT)and mean platelet volume(MPV),P-selectin,nitric oxide(NO)in whole blood(WB),the levels of interleukin-1β(IL-1β)and tumor necrosis factor-a(TNF-α)in colonic tissue.Results After gavage,PLT was increased(P<0.05),while MPV was decreased(P<0.05)in the model group relevant to the control group;lower PLT in both Ershen Pill groups,and higher MPV in the low-dose Ershen Pill group were detected than those in the model group(all P<0.05);PLT,MPV were found no significant difference from the positive drug group vs the high-dose Ershen Pill group vs the model group(P>0.05),the difference was not statistically significant in PLT,MPV between the low-and high-dose Ershen Pill groups(P>0.05).P-selectin and NO were increased in the model group than in the control group(P<0.05);NO in the positive drug group,both Ershen Pill groups,and P-selectin in the high-dose Ershen Pill group were decreased relevant to the control group(P<0.05);there was no significant difference in P-selectin between the positive drug group,the low-dose Ershen Pill group and the model group(P>0.05);no difference in the P-selectin and NO between both Ershen Pill groups(P>0.05).IL-1βand TNF-αof colonic tissue in the model group were higher than those in the control group(P<0.05),which in the positiv
