摘  要:目的 采用超高效色谱-电喷雾/四级杆飞行时间串联质谱联用(UPLC-ESI-Q-TOF-MS/MS)技术对三七粉中主要化学成分和血中移行成分进行准确快速鉴定。方法 采用Waters Xevo G2 Q-Tof高分辨质谱、Waters Acquity UPLC液相色谱系统、Waters Acquity UPLC?BEH C18色谱柱(2.1×100 mm,1.7μm),以0.1%甲酸乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,采用电喷雾(ESI)离子源,在正、负离子模式下采集数据,通过与对照品的保留时间及质谱信息对比,并结合文献报道及质谱数据库中相同成分或同类成分的精确分子质量、质谱裂解规律进行匹配,对三七粉中化学成分和血中移行成分进行快速准确识别。结果三七粉中共鉴定得到54个化学成分,包括原人参二醇皂苷类36个,原人参三醇皂苷类14个,黄酮类1个,糖类1个,氨基酸类1个和酯类化合物1个。在给药大鼠血浆中共鉴定得到18个三七粉相关的外源性化合物,包括14个吸收原型药物成分和4个代谢物。结论 利用建立的定性分析方法快速准确地识别三七粉中主要化学成分和血中移行成分。明确了三七粉体外化学成分组-体内血行成分组的药效物质基础传递-转化过程,为三七粉的质量标志物确定与质量控制提供依据,也为其深入分子作用机制研究奠定基础。Objective To establish a method to recognize and classify the main chemical constituents and constituents absorbed into blood of Radix Notoginseng Powder accurately and rapidly by applying novel ultra-performance liquid chromatography tandem quadrupole time of flight mass spectrometry(UPLC-Q-TOF-MS/MS). Methods ACQUITY UPLC? BEH C18 chromatographic column(100 mm × 2.1 mm, 1.7 μm) was employed to UPLC analysis with the mobile phase consisting of 0.1% formic acid acetonitrile-0.1% formic acid aqueous solution. ESI ion source was used to ensure the data collected in positive and negative ion mode. The chemical components in Radix Notoginseng powder and the transitional components in blood were quickly and accurately identified by comparing the retention time and mass spectrometry information with the reference substance, combined with the literature reports and the accurate molecular weight and mass spectrometry cracking law of the same or similar components in the mass spectrometry database.Results A total of 54 chemical components were identified, including 36 protopanaxadiol saponins, 14 protopanaxatriol saponins, 1 flavonoid, 1 carbohydrate, 1 amino acid and 1 ester. A total of 18 exogenous compounds related to Radix Notoginseng Powder were identified in the plasma of rats, including 14 absorbed prototype components and 4metabolites. Conclusion The qualitative method established in this study could be used to rapidly and accurately identify the chemical constituents and constituents absorbed into blood of Radix Notoginseng Powder. This study clarified the transfer-transformation process of pharmacodynamic substances from in vivo to blood, and provided the basis for the determination of the quality marker and quality control of the Radix Notoginseng Powder. Lay the foundation for molecular mechanism research deeply.
