摘  要:旨在探究山羊卵巢黄体生命周期中细胞自噬相关信号通路分子的表达。分别采集山羊早期(E)、中期(M)和晚期(L)黄体的卵巢,应用蛋白免疫印迹(WB)、免疫组织化学(IHC)技术检测LC3B、JAK2、p-JAK2、STAT3、p-STAT3在黄体组织中的表达与定位。WB结果显示:LC3B的表达水平在黄体晚期显著升高(P<0.05);黄体早期p-JAK2的表达水平极显著高于中期和晚期(P<0.01),JAK2表达水平在黄体不同发育时期无显著差异(P>0.05);p-STAT3和STAT3表达水平在黄体不同发育时期均无显著差异(P>0.05)。IHC结果显示:LC3B、STAT3主要定位于黄体类固醇生成细胞的细胞质中,p-STAT3主要定位在黄体类固醇生成细胞的细胞核中。以上结果表明可能存在多条信号通路调控山羊黄体细胞自噬,为深入探究山羊黄体形成与退化的分子机制提供科学依据。This research was to study the expression of autophagy-related signaling molecules in the lifespan of the corpus luteum in the goat ovary.Ovary samples of the early(E),middle(M)and late(L)luteal phases were collected from goats,and the luteal localization and expression of the LC3B protein and its associated JAK2/STAT3 signaling pathway were detected by Western blot(WB)and immunohistochemistry(IHC).The WB results showed that the expression level of LC3B was significantly higher in the late luteal phase(P<0.05);the expression level of p-JAK2 in the early luteal phase was very significantly higher than that in the mid and late luteal phases(P<0.01),but the expression level of JAK2 was not significantly different in different developmental periods of the corpus luteum(P>0.05);and the expression level of neither p-STAT3 nor STAT3 showed any significance in the different luteal periods(P>0.05).The IHC results showed that LC3B and STAT3 were mainly localized in the cytoplasm of the steroidogenic cells,and p-STAT3 was mainly localized in the nucleus of these cells.In conclusion,these results suggested that there might be multiple signaling pathways regulating autophagy in the corpus luteum cells of goats,which provided a scientific basis for in-depth investigation of the molecular mechanism of corpus luteum formation and degeneration in goats.
