摘  要:为调查广西地区某规模化猪场疑似患猪传染性胸膜肺炎猪的病原、血清型及有效治疗药物等情况,首先对患病猪进行剖检观察其病理变化,同时无菌采集病猪肺脏等病料,接种平板后纯化培养,然后通过革兰染色镜检、生化鉴定、16S rRNA序列比对、血清型及毒素基因分析等方法进行鉴定,并对分离菌株的耐药性和动物致病性进行探究。结果:从病料中分离到1株细菌,其菌落、菌体形态、生理生化特性和16S rRNA基因序列符合胸膜肺炎放线杆菌(APP)特征,该菌株含有ApxⅠ、ApxⅡ、ApxⅣ三种毒素,属于APP血清5型,因此将其命名为APP5-YXLM;药敏试验结果显示,该菌对β-内酰胺类药物和头孢类药物敏感,对红霉素、庆大霉素、多西环素、四环素等6种药物耐药;动物致病性试验表明,该菌株对小鼠具有较强的致病性,其半数致死量(LD_(50))为7.06×10^(8) CFU。试验结果为该发病猪场猪传染性胸膜肺炎的防治提供了理论依据,同时也为掌握胸膜肺炎放线杆菌在广西地区的流行情况提供了数据支持。In order to identify the pathogen species,serotypes and effective treatment antibiotics of pigs suspected of porcine contagious pleuropneumonia at a large-scale pig farm in the Guangxi area.the lung samples of diseased pigs used in this study were collected aseptically.The samples received purifying culture after plate inoculation and were tested by Gram stain microscopic examination,biochemical identification,16S rRNA sequence comparison,and serotype and toxin gene analysis.Finally,the antibiotics resistance and animal pathogenicity of the isolated strains were explored.The results showed that a strain of Actinobacillus pleuropneumoniae was isolated from the diseased pig lungs,and the colony,morphology,physiological,biochemical characteristics and 16S rRNA gene sequence of the strain were consistent with the characteristics of Actinobacillus pleuropneumoniae.It contained three toxins:ApxⅠ,ApxⅡ,and ApxⅣ,belonging to the APP serotype 5 strain;therefore,it was named APP5-YXLM.The antibiotics susceptibility test showed that the bacterium was sensitive toβ-lactam antibiotics and cephalosporins antibiotics,and was resistant to six antibiotics including erythromycin,gentamicin,doxycycline and tetracycline,and so on.The animal pathogenicity test showed that APP5-YXLM had strong pathogenicity in mice with an LD_(50) of 7.06×10^(8) CFU The results of this experiment provided a theoretical basis for prevention and treatment of porcine contagious pleuropneumonia in pig farms,and also supplied data support for future research on the prevalence of Actinobacillus pleuropneumoniae in the Guangxi area.
