摘 要:目的:探讨香青兰总黄酮(total flavones of Dracocephalum moldavica L.,TFDM)对帕金森病(Parkinson’s disease,PD)模型小鼠中脑黑质内多巴胺转运蛋白表达的效果。方法:C517BL/6J小鼠腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)制备PD动物模型,多种行为学实验方法评价动物运动功能;尼氏染色观测黑质区多巴胺能神经元的数量;RT-PCR和Western blot检测黑质酪氨酸羟化酶(tyrosine hydroxylase,TH)、囊泡单胺转运蛋白2(Vesicular monoamine transporter 2,VMAT2)和多巴胺转运蛋白(Dopamine transporter,DAT)的表达情况。结果:行为学检测表明,TFDM组显著增加小鼠的运动距离(P<0.05),显著增加小鼠的下落潜伏期(P<0.05),明显减少小鼠的爬杆时间(P<0.05);MPTP腹腔注射明显减少了黑质内多巴胺能神经元的数量,TFDM显著的提高了PD模型小鼠黑质内多巴胺能神经元数量;RT-PCR结果显示,TFDM组的TH、VMAT2和DAT的mRNA表达均显著增高(P<0.05);TFDM组TH、VMAT2和DAT蛋白表达均明显上调(P<0.05)。结论:TFDM可能通过上调VMAT2和DAT蛋白的表达,减少DA能神经元丢失,进而改善PD模型小鼠运动功能障碍。Objective:To explore the effect of total flavones of Dracocephalum moldavica L.(TFDM)on the expression of dopamine transporter in the substantia nigra of Parkinson's disease(PD)model mice.Methods:C57BL/6J mice were intraperitoneal injection with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)to prepare a PD animal model,and a variety of behavioral experimental methods were used to evaluate animal motor function.Nissl staining was used to observe the number of dopaminergic neurons in the substantia nigra.RT-PCR and Western blot were used to test tyrosine hydroxylase(TH),vesicular monoamine transporter 2(VMAT2)and dopamine transporters(DAT).Results:Behavioral tests showed that the TFDM group significantly increased the movement distance of mice(P<0.05),significantly increased the falling latency of mice(P<0.05),and significantly reduced the climbing time of mice(P<0.05).MPTP intraperitoneal injection significantly reduced the number of dopaminergic neurons in substantia nigra,and TFDM significantly increased the number of dopaminergic neurons in substantia nigra of PD model mice.RT-PCR results showed that the mRNA expressions of TH,VMAT2 and DAT in TFDM group were significantly increased(P<0.05).The expressions of TH,VMAT2 and DAT proteins in TFDM group were significantly up-regulated(P<0.05).Conclusion:TFDM may reduce the loss of dopaminergic neurons by up-regulating the protein expression levels of VMAT2 and DAT,thereby improving motor dysfunction in PD model mice.
