摘 要:目的通过相关指标的检测,探究园参茎叶总皂苷和林下山参茎叶总皂苷对气虚血瘀模型大鼠的作用差异,为不同生境人参茎叶资源的开发利用提供理论依据。方法采用游泳力竭联合肾上腺素皮下注射法,构建气虚血瘀模型大鼠。分别给予不同剂量(10、20、40 mg/kg)园参茎叶总皂苷和林下山参茎叶总皂苷干预后,经腹主动脉取血,进行血液流变学检测;通过酶联免疫吸附法检测肌酸激酶(CK)、肌酸激酶同工酶MB(CK-MB)、内皮素-1(ET-1)、一氧化氮(NO)、血栓素B2(TXB2).Na^(+)-K^(+)-ATP酶的表达。结果与正常组相比,模型组的各项指标均有显著升高或降低,具有统计学意义(P<0.01)。与模型组相比,园参茎叶总皂苷和林下山参茎叶总皂苷给药对降低气虚血瘀模型大鼠血清CK CK-MB.ET-1.TXB2水平及全血黏度高切、中切、低切水平.提高气虚血瘀模型大鼠血清NO.Na_(+)-K^(+)-ATP酶水平均具有--定的作用,并且具有统计学差异(P<005,P<0.01)。而园参茎叶总皂苷与林下山参茎叶总皂苷对气虚血瘀模型大鼠的改善作用未见统计学差异(P>0.05)。结论园参茎叶总皂苷和林下山参茎叶总皂苷均能改善气虚血瘀模型大鼠的血液流变学指标,其作用机制可能与保护心肌细胞.改善内皮功能、保证能量代谢等有关;且同等剂量园参茎叶总皂苷与林下山参茎叶总皂苷对气虚血瘀模型大鼠的作用相当。Objective Through the comparative study on the effects of total saponins from stems and leaves of garden ginseng(G-GS)and ginseng under forest(F-GS)on Qi-deficiency and blood-stasis syndrome in rats to provide theoretical basis for the development and utilization of stem and leaf resources of Panax ginseng in different habitats.Methods Qi-deficiency and blood-stasis syndrome in rats were established by swimming exhaustion combined with subcutaneous injection of epinephrine.After treatment with different doses(10,20,40 mg/kg)of G-GS and F-GS,blood was collected from the abdominal aorta for further hemorheology detection.The expression of creatine kinase(CK),creatine kinase isoenzyme MB(CK-MB),endothelin-1(ET-1),nitric oxide(NO),thromboxane B2(TXB2)and Na^(+)-K^(+)-ATPase were detected by enzyme-linked immunosorption.Results Compared with the normal group,a message is displayed indicating that the model was successfully established(P<0.01).Compared with the model group,administration of G-GS and F-GS could reduce the levels of CK,CK-MB,ET-1,TXB2 in serum and the high,medium and low levels of whole blood viscosity on Qi-deficiency and blood-stasis syndrome in rats,while the serum levels of NO and Na^(+)-K^(+)-ATPase were increased,with statistical significance(P<0.05,P<0.01).There was no statistical difference between G-GS and F-GS on the improvement of Qi-deficiency and blood-stasis syndrome in rats(P>0.05).Conclusion G-GS and F-GS all can improve the hemorheology indexes on Qi-deficiency and blood-stasis syndrome in rats,and the mechanism may be through protecting myocardial cells,improving endothelial function and ensuring energy metabolism.The effect of G-GS and F-GS on Qi-deficiency and blood-stasis syndrome in rats was similar at the same dose.
