摘 要:目的观察电针手厥阴心包经穴对大脑中动脉栓塞(MCAO)模型大鼠突触可塑性的影响,探讨其作用机制。方法将150只SD大鼠随机分为正常组、假手术组、模型组、心包经组和肺经组,每组再分成3、7、14、21、28 d 5个时点,每个时点6只。采用颈外动脉插入线栓法制备MCAO大鼠模型。心包经组取穴“天泉”“曲泽”“内关”“大陵”,肺经组取穴“天府”“尺泽”“列缺”“太渊”,于造模后第2日开始干预,每次30 min,连续电针6 d,间隔1 d,继续电针,共28 d。采用免疫组化、Western blot检测大鼠脑组织突触素(SYP)、突触后致密蛋白-95(PSD-95)表达。结果免疫组化结果显示,与正常组、假手术组比较,模型组大鼠脑组织7、14 d SYP表达明显降低(P<0.05,P<0.01),各时点PSD-95表达均明显降低(P<0.01);与模型组比较,心包经组、肺经组大鼠脑组织各时点SYP表达均明显升高(P<0.01,P<0.05),除3 d外,心包经组、肺经组大鼠脑组织PSD-95表达均明显升高(P<0.01);与同一时点肺经组比较,除3 d外,心包经组大鼠脑组织SYP和PSD-95表达均明显升高(P<0.01,P<0.05)。Western blot结果显示,与正常组、假手术组比较,模型组大鼠脑组织各时点SYP和PSD-95蛋白表达均明显降低(P<0.01);与模型组比较,心包经组大鼠脑组织各时点SYP和PSD-95蛋白表达均明显升高(P<0.01,P<0.05);与同一时点肺经组比较,心包经组大鼠脑组织3、7、21 d SYP和PSD-95蛋白表达均明显升高(P<0.01)。结论电针心包经穴能提高MCAO模型大鼠突触可塑性相关蛋白SYP和PSD-95表达,在一定程度上参与脑缺血后突触可塑性的调节。Objective To observe the effects of electroacupuncture on the acupoints of the pericardium meridian on the synaptic plasticity of middle cerebral artery occlusion(MCAO)model rats;To discuss its mechanism of action.Methods Totally 150 SD rats were randomly divided into normal group,sham-operation group,model group,pericardial meridian group,and lung meridian group,each group was divided into 3,7,14,21,28 d time points,6 for each time point.The external carotid artery was used to insert a suture to create MCAO rat model.“Tianquan”,“Quze”,“Neiguan”and“Daling”were selected as the acupoints of the pericardium meridian group,and“Tianfu”,“Chize”,“Lieque”and“Taiyuan”were selected as the acupoints of the lung meridian group.The treatment was started on the second day after modeling,30 minutes each time,continuous electroacupuncture for 6 days,1 day apart,continued with electroacupuncture for 28 d.Immunohistochemistry and Western blot were used to detect the expressions of SYP and PSD-95 in brain tissue.Results Immunohistochemistry results showed that compared with the normal group and the sham-operation group,the expression of SYP in brain tissue of the rats in model group decreased at 7 and 14 days(P<0.05,P<0.01),the expression of PSD-95 significantly decreased at each time point(P<0.01).Compared with the model group,the expression of SYP in brain tissue of rats in the pericardium meridian group and lung meridian group significantly increased at each time point(P<0.01,P<0.05),the expression of PSD-95 in brain tissue of rats in the pericardial meridian group and lung meridian group increased significantly(P<0.01)except for the 3rd day.Compared with the lung meridian group at the same time point,the expressions of SYP and PSD-95 in brain tissue of rats in the pericardial meridian group increased significantly(P<0.01)except for the 3rd day.Western blot results showed that compared with the normal group and the sham-operation group,the proteins expression of SYP and PSD-95 in brain tissue of r
