摘 要:目的建立射干药材中总三萜酸含量测定方法,并对测定结果进行不确定度分析。方法以熊果酸为对照品,采用香草醛高氯酸系统测定总三萜酸含量;根据测定方法的数学模型,对测定过程中引入的不确定度进行分析评估,并用扩展不确定度表示不同产地射干总三萜酸含量测定结果。结果射干总三萜酸含量在18.42~92.08μg范围内线性关系良好(r=0.9992),平均加样回收率为98.44%;测定方法的不确定度主要来源于标准曲线拟合。25批射干药材中总三萜酸含量为32.83~66.65 mg/g,相应扩展不确定度为2.18~4.30 mg/g。结论本研究建立的射干药材中总三萜酸含量测定方法重复性、准确度良好,用扩展不确定度表达射干中总三萜酸含量更为合理,可用于射干药材质量控制。Objective To establish a content determination method for total triterpenes in Belamcandae Rhizoma;To carry out the uncertainty analysis of the measurement results.Methods The content of total triterpenoidic acid was determined by vanillin-perchloric acid chromogenic system with ursolic acid as reference;according to the mathematical model of the determination method,the uncertainty introduced in the determination process was analyzed and evaluated,and the expanded uncertainty was used to express the determination results of the total triterpene acid content of Belamcandae Rhizoma from different producing places.Results The linear range of total triterpenoid acid content was 18.42–92.08μg(r=0.9992).The average recovery was 98.44%.The evaluation results showed that the uncertainty mainly comes from the curve calibration fitting.The content of total triterpenoid acid in 25 batches of samples was between 32.83 mg/g and 66.65 mg/g,and the corresponding expanded uncertainty ranged from 2.18 mg/g to 4.30 mg/g.Conclusion The content determination method established in this study is reproducible and accurate.It is more reasonable to express the content of total triterpene acid in Belamcandae Rhizoma with the expanded uncertainty,which can be used for quality control of Belamcandae Rhizoma medicinal materials.
