摘  要:该研究利用cDNA末端快速扩增(RACE)技术成功克隆到大鳍鳠的GHR基因,获得了其完整的CDS区序列;通过同源序列比对构建了大鳍鳠的系统进化树,并采用生物信息学的方法分析了大鳍鳠GHR蛋白的特性。结果表明:大鳍鳠GHR基因CDS区长度为1731 bp,编码576个氨基酸;大鳍鳠与黄颡鱼、斑点叉尾鮰的亲缘关系较近,它们之间GHR基因的同源性分别为90.3%、88.7%,但是与哺乳动物的亲缘关系较远;大鳍鳠GHR蛋白分子式为C_(2900)H_(4483)N_(765)O_(888)S_(28),属于跨膜蛋白,其二级结构由α-螺旋(20.66%)、延伸链(24.13%)和无规则卷曲(55.21%)组成,三级结构预测结果与二级结构一致;该蛋白能与GH1、GH2、CISH、JAK2和STAT5A等分子形成互作网络。Growth hormone receptor(GHR)gene from the Hemibagrus macropterus genome was cloned using rapid-amplification of cDNA ends(RACE).The complete CDS region of the gene was obtained and bioinformatics analysis related to the gene was performed.The results showed that the CDS region of GHR gene contained 1731bp and encoded 576 amino acids.The sequence was similar to those from Pelteobagrus fulvidraco and Ictalurus punctatus(90.3% and 88.7%,respectively).H.macropterus as fish was distantly related to mammals.The molecular formula of GHR protein is C_(2900)H_(4483)N_(765)O_(888)S_(28),which belongs to transmembrane protein.GHR protein in the secondary structure is composed ofα-helix(20.66%),extended chain(24.13%)and random curl(55.21%),and consistent with the prediction result of tertiary structure.The protein can form an interaction network with GH1,GH2,CISH,JAK2 and STAT5A.These results provide theoretical reference for further study of function and related signaling path ways on H.macropterus GHR gene.
