摘 要:目的:优化大黄总蒽醌的醇提工艺。方法:采用渗漉法提取大黄总蒽醌,应用HPLC测定其含量并计算大黄总蒽醌提取率。影响渗漉效果的主要因素有乙醇浓度、乙醇用量、渗漉速度,因此选择上述3个因素作为考察因素,按L^(9)(3^(4))的试验安排进行提取,以正交试验的9组数据作为训练样本,应用误差反向传播人工神经网络(BP⁃ANN)进行建模及分析,以大黄总蒽醌提取率作为考察指标,仿真模拟预测大黄总蒽醌最佳醇提工艺参数。结果:正交试验优化的提取工艺为以60%乙醇浸泡24 h,以1 ml·(min·kg)^(-1)的渗漉速度收集10倍量渗漉液,大黄总蒽醌提取率为2.142%;BP⁃ANN预测的最佳提取工艺为70%乙醇浸泡24 h,以3 ml·(min·kg)^(-1)的渗漉速度收集10倍量渗漉液,大黄总蒽醌提取率为2.380%。验证试验结果表明,BP⁃ANN优化工艺与正交试验相比,大黄总蒽醌提取率的的增长率为8.03%,与预测值的相对误差为2.77%。结论:BP⁃ANN优化工艺的提取效果优于正交试验。Objective:To optimize the extract process of five anthraquinones from Rhei Radix et Rhizoma.Methods:Five anthraquinones in rhubarb were extracted by percolation and determined by HPLC.For the percolation method,the main factors affecting the extraction effect were ethanol concentration,ethanol amount and percolation speed,which were selected as the investigation factors,and the extraction conditions were optimized according to L^(9)(3^(4))orthogonal test.The content of five anthraquinones in the extraction solution was taken as the investigation index.The stable and feasible extraction process of rhubarb was screened through visual analysis and variance analysis of SPSS13.0,and then back propagation artificial neural network(BP-ANN)was used to further predict the optimum extraction parameters.Results:The extraction process optimized by orthogonal test was as follows:soaking in 60%ethanol for 24 hours and collecting 10-fold amount of percolate at the percolation rate of 1 ml·(min·kg)^(-1).The extraction rate of total anthraquinones in rhubarb was 2.142%;BP-ANN predicted that the best extraction process was to soak in 70%ethanol for 24 hours and collect 10-fold amount of percolation solution at the percolation rate of 3 ml·(min·kg)^(-1).The extraction rate of total anthraquinones in rhubarb was 2.380%.The results of validation test showed that compared with the orthogonal test,the extraction increase rate of total anthraquinones of rhubarb was 8.03%,and the relative error with the predicted value was 2.77%.Conclusion:BP-ANN prediction is better than orthogonal test in optimizing extraction process to improve extraction effect.
