摘 要:目的 建立糖尿病小鼠模型并探讨辅酶Q_(10)对糖尿病小鼠肝肾损伤以及骨骼的影响。方法 除正常组(n=20)外,其余小鼠通过腹腔注射50 mg·kg^(-1)链脲佐菌素建立糖尿病小鼠模型,造模后随机分为3组:模型组(n=20)、二甲双胍组(n=10)和辅酶Q_(10)组(n=10)。正常组和模型组小鼠灌胃生理盐水,二甲双胍组灌胃500 mg·kg^(-1)盐酸二甲双胍,辅酶Q_(10)组灌胃3 000 mg·kg^(-1)辅酶Q_(10)。各组小鼠每天灌胃给药1次,连续75 d。给药期间定期测定小鼠空腹血糖。给药结束后,用分光光度计与酶标仪检测血清生化指标水平;用三点弯曲实验测定骨生物力学参数;用苏木精-伊红染色法与糖原染色法观察小鼠肾、肝组织病理学变化。结果 正常组、模型组、二甲双胍组和辅酶Q_(10)组小鼠肾功能指标肌酸酐(Cr)含量分别为(152.23±40.08),(390.60±85.32),(346.64±37.20)和(277.27±62.36)μmol·L^(-1),模型组与正常组相比以及辅酶Q_(10)组与模型组相比,差异均有统计学意义(均P<0.05);这4组小鼠肝功能指标谷丙转氨酶分别为(65.97±12.09),(128.92±20.01),(95.40±13.36)和(100.16±9.52)U·L^(-1),模型组与正常组比较,辅酶Q_(10)组与模型组比较,差异均有统计学意义(均P<0.05);这4组小鼠骨生物力学参数断裂载荷分别为(30.83±4.90),(24.87±6.84),(31.05±6.31)和(25.90±4.20)N,模型组与正常组相比,差异均有统计学意义(均P<0.05),辅酶Q_(10)组与模型组相比,断裂载荷有升高趋势,但差异无统计学意义(P>0.05)。结论 辅酶Q_(10)改善糖尿病小鼠肝、肾损伤,对骨骼的硬度可能有积极的作用。Objective To establish diabetic mouse model and investigate the effects of coenzyme Q_(10)on liver and kidney injury and bone in diabetic mice. Methods Except for normal group(n=20), other mice were intraperitoneally injected with streptozotocin 50 mg·kg^(-1)to establish a diabetic mouse model, and were randomly divided into three groups: model group(n=20), metformin group(n=10), coenzyme Q_(10)group(n=10). Mice in normal group and model group were given normal saline, mice in metformin group were given 500 mg·kg^(-1)metformin hydrochloride, mice in coenzyme Q_(10)group were given 3 000 mg·kg^(-1)coenzyme Q_(10). All mice were intragastric once a day for 75 days. During the experiment, the body weight of the mice was regularly recorded and fasting blood glucose was measured. After administration, serum biochemical indexes were detected by spectrophotometer and enzyme reader. The biomechanical parameters of bone were determined by three-point bending test. The histopathological changes of kidney and liver were observed by Htoxylin-eosin stain and Periodic Acid-Schiff stain.
