莱菔子水煎液降低自发性高血压大鼠血压的粪便代谢组学研究

摘　　要：目的考察莱菔子水煎液对自发性高血压大鼠(SHR)的降血压作用及其对SHR粪便代谢物的影响,分析特征代谢物可能涉及的代谢通路,为探索莱菔子降血压的机制提供实验依据。方法选用8周龄雄性SHR,将其随机分为模型组和莱菔子水煎液组,并用8周龄雄性Wistar-Kyoto(WKY)大鼠作对照,灌胃6周后取大鼠粪便,利用液相色谱-质谱法采集粪便代谢物数据,利用SIMCA-P 14.0、MetaboAnalyst 4.0等软件对数据进行分析。结果莱菔子水煎液对SHR有明显的降血压作用(P<0.05),并调节了SHR粪便中亚油酸、尿香草苦杏仁酸、鹅去氧胆酸等代谢物的水平,其相关代谢通路为亚油酸代谢通路、苯丙氨酸代谢通路、卟啉与叶绿素代谢通路、花生四烯酸代谢通路等。结论莱菔子水煎液具有确切的降血压作用,并可改变SHR内源性代谢物。Objective To investigate the hypotensive effect of Radish seed decoction on spontaneously hypertensive rats(SHR)and its effect on fecal metabolites of SHR,to analyze the possible metabolic pathways involved in the characteristic metabolites,and to provide an experimental basis for exploring the mechanism of blood pressure lowering by radish seed.Methods Eight-week-old male SHR were randomly divided into model group and radish seed decoction group,and 8-week-old male WKY rats were used as control.Feces were collected after 6 weeks of gavage.The data of fecal metabolites were collected by untargeted liquid chromatography-mass spectrometry and analyzed by SIMCA-P 14.0,MetaboAnalyst 4.0 and other software.Results Compared with the model group,Radish seed decoction had a significant hypotensive effect on SHR(P<0.05)and modulated the levels of fecal metabolites such as linoleic acid,Vanillylmandelic acid and Chenodeoxycholic acid.The related metabolic pathways are linoleic acid metabolic pathway,phenylalanine metabolic pathway,porphyrin and chlorophyll metabolic pathway,and arachidonic acid metabolic pathway.Conclusion Radish seed decoction has a definite hypotensive effect and modifies the endogenous metabolites of SHR.
