摘 要:目的:观察茵陈四苓颗粒联合骨髓间充质干细胞(BMSCs)对急性肝衰竭大鼠线粒体膜电位的调节作用。方法:将60只雄性SPF级SD大鼠随机分成A组(正常组)、B组(茵陈四苓颗粒联合骨髓间充质干细胞组)、C组(茵陈四苓颗粒组)、D组(模型组)、E组(复方甘草酸苷组)、F组(骨髓间充质干细胞组),每组各10只。采用硫代乙酰胺腹腔注射的方法制备急性肝衰竭大鼠模型,进行BMSCs的分离纯化、培养及鉴定,观察大鼠的一般情况与存活情况,血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总胆红素(TBil)的含量,肝脏组织病理情况、肝线粒体的形态与结构、肝线粒体膜电位。结果:大鼠血清ALT、AST、TBil水平比较,D组较A组明显升高(P<0.05),B、C、E、F组较D组降低,其中B组降低更显著,差异均有统计学意义(P<0.05),E、F组组间差异无统计学意义(P>0.05)。B、C、E、F组肝脏组织病理、线粒体形态与结构的变化情况较D组好转。D组Q1-UR/Q1-LR阳性百分比比值显著升高,与A组比较,差异有统计学意义(P<0.05);B、C组与D组比较,差异有统计学意义(P<0.05);各干预组肝线粒体膜电位稳定,E组<F组<C组<B组,组间比较,差异均有统计学意义(P<0.05)。结论:茵陈四苓颗粒联合骨髓间充质干细胞移植对急性肝衰竭大鼠线粒体膜电位发挥调节作用,并通过抑制线粒体凋亡来改善急性肝衰竭,保护肝细胞。Objective: To investigate the regulatory effect of Yinchen Siling granules combined with bone marrow mesenchymal stem cells(BMSCs) on mitochondrial membrane potential in rats with acute liver failure.Methods: A total of 60 specific pathogen-free male Sprague-Dawley rats were randomly divided into group A(normal group),group B(Yinchen Siling granules+BMSCs group),group C(Yinchen Siling granules group),group D(model group),group E(compound glycyrrhizin group),and group F(BMSCs group),with 10 rats in each group.Intraperitoneal injection of thioacetamide was performed to establish a rat model of acute liver failure, and BMSCs were isolated, purified, culture, and identified.The rats were observed in terms of general status, survival, serum content of aspartate aminotransferase(AST)/alanine aminotransferase(ALT)/total bilirubin(TBil),liver histopathology, liver mitochondrial morphology and structure, and liver mitochondrial membrane potential.Results: Compared with group A,group D had significantly higher serum levels of ALT,AST,and TBil(P<0.05);compared with group D,groups B,C,E,and F had significantly lower serum levels of ALT,AST,and TBil, and group B had significantly greater reductions than the other groups(P<0.05);there were no significant differences in these indices between group E and group F(P>0.05).Compared with group D,groups B,C,E,and F had significant improvements in liver histopathology and liver mitochondrial morphology and structure.Compared with group A,group D had a significant increase in the percentage of Q1-UR/Q1-LR-positive cells(P<0.05),and there was also a significant difference between group B/C and group D(P<0.05).Each intervention group had a stable liver mitochondrial membrane potential, and group B had the highest potential, followed by groups C,F,and E(P<0.05).Conclusion: Yinchen Siling granules combined with BMSCs exert a regulatory effect on mitochondrial membrane potential in rats with acute liver failure and can improve acute liver failure and protect the liver by inhibiting mitochon
