摘 要:TCP(Teosinte branched 1/Cycloidea/Proliferating cell factors)转录因子作为植物特有的一类转录因子,参与植物生长发育的多个重要进程。本研究利用生物信息学方法对花生TCP基因家族成员进行鉴定,结合花生侧枝转录组数据进行表达分析。结果表明:在四倍体花生AABB、二倍体AA和BB基因组中分别鉴定获得30个AhTCPs、11个AduTCPs和10个AipTCPs,定位到14、6和5条染色体上;与拟南芥AtTCPs蛋白聚类分析发现,花生TCP蛋白可分为ClassⅠ和ClassⅡ(CYC/TB1和CIN)2个亚类;AhTCPs基因中含有1~6个数量不等的外显子和5个不同的motif;蛋白质长度为131~658个氨基酸,分子量为9.94~72.66 kD;大部分成员定位在细胞核上;家族基因启动子顺式作用元件分析发现,在AA、BB、AABB基因组分别获得77、75和94种顺式作用元件。转录组结果分析表明,24个AhTCPs在侧枝发育过程中存在不同程度的表达,大多数AhTCPs基因在2个品种(‘冀花5号’和‘M130’)6个时期中高表达。以上结果为进一步了解花生TCP家族的生物学功能提供理论基础。TCP(Teosinte branched 1/Cycloidea/Proliferating cell factors) genes are plant unique transcription factors involving in many important processes of growth and development. In the present study, members of peanut members of TCP gene family was identified followed by bioinformatics analysis of their chromosome location, phylogenetic evolution, prediction of physical and chemical properties, promoter cis elements and genome collinearity. The results showed that 30 AhTCPs, 11 AduTCPs and 10 AipTCPs were located on 14, 6 and 5 chromosomes in the tetraploid genome and the wild diploid genomes(AA and BB), respectively. These peanut TCP genes clustered into two groups in the phylogenetic tree. AhTCP genes contained 1-6 exons including 5 different motifs. The length of proteins was 131-658 aa and the molecular weight was 9.94-72.66 kD. Most members were located in the nucleus. A total of 77,75 and 94 cis-acting elements of family gene promoters were obtained in AA, BB and AABB genomes, respectively.The results of transcriptome analysis showed that most AhTCP genes were highly expressed in 6 stages of developing lateral branches with different patterns in ’Jihua 5’ and ’M130’. These results will provide theoretical basis for further understanding the biological function of TCP family in peanut.
