玉米转录因子ZmMYC7对ZmLOX9的调控作用研究

摘  要:为探究玉米茉莉酸(JA)信号通路中抗病关键转录因子ZmMYC7与其候选靶基因ZmLOX9的关系,本研究利用生物信息学方法对ZmLOX9的系统进化关系和启动子元件进行了分析,利用qRT-PCR技术检测了ZmMYC7基因突变对ZmLOX9基因表达的影响,并利用酵母单杂交(Y1H)技术检测了ZmMYC7对ZmLOX9启动子的结合互作。结果发现,玉米ZmLOX9与高粱SbLOX6、谷子SiLOX6等同源,ZmLOX9启动子区域含有ZmMYC7的作用元件G-box;ZmMYC7基因的突变体接种禾谷镰孢(Fusarium graminearum)后,ZmLOX9表达量显著下调;Y1H试验发现,ZmMYC7能够与ZmLOX9启动子作用,且ZmLOX9启动子区G-box是其重要作用位点。本研究结果明确了转录因子ZmMYC7直接作用于ZmLOX9启动子区G-box进而调控ZmLOX9基因的表达,为阐明转录因子ZmMYC7调控玉米抵抗病菌侵染的机制奠定了基础。ZmMYC7 is a key transcription factor in maize JA signaling pathway regulating disease resistance.In order to explore the relationship between ZmMYC7 and its candidate target ZmLOX9, we first analyzed the phylogenetic evolution and cis-elements of ZmLOX9 promoter using bioinformatics methods. Then detected the expression of ZmLOX9 in ZmMYC7 mutants by qRT-PCR and interaction of ZmMYC7 with ZmLOX9 promoter by Y1H assay. The results showed that ZmLOX9 was close to SbLOX6 of Sorghum bicolor and its promoter contains the G-box element. ZmLOX9 expression was significantly lower in the ZmMYC7 EMS mutants after inoculated by Fusarium graminearum. Y1H assay showed that ZmMYC7 bound directly to the G-box region of Zm LOX9 promoter.This study clarified that transcription factor ZmMYC7 directly recognize the G-box of ZmLOX9 promoter to regulate its expression, which laid a foundation for clarifying the mechanism of ZmMYC7 in regulating maize resistance to pathogen infection.
