摘  要:目的探究长链非编码RNA(LncRNA)小核仁RNA宿主基因17(SNHG17)在气道平滑肌细胞(ASMCs)中的表达及其作用。方法腹腔注射卵清蛋白抗原悬浮液构建幼龄Wistar大鼠的支气管哮喘模型。将大鼠随机分为对照组(腹腔注射生理盐水)和模型组(腹腔注射10%的卵清蛋白抗原悬浮液),随后将空白质粒(pc-NC组)、SNHG17过表达质粒(pc-SNHG17组)分别转染至模型组ASMCs中。用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测ASMCs中SNHG17和miR-375表达水平,以EdU法检测ASMCs增殖能力,以划痕实验检测ASMCs迁移能力。结果对照组和模型组中SNHG17的表达水平分别为1.00±0.10和1.81±0.11。对照组、模型组、pc-NC组和pc-SNHG17组中miR-375的表达水平分别为1.01±0.08,0.72±0.05,0.79±0.05和0.53±0.06;细胞增殖率分别为(26.11±2.51)%,(33.01±2.91)%,(34.44±3.07)%和(43.92±2.97)%;细胞迁移率分别为(10.05±1.00)%,(18.62±1.20)%,(20.03±1.12)%和(44.71±3.33)%。以上指标,模型组与对照组相比,pc-SNHG17组与pc-NC组相比,差异均有统计学意义(均P<0.05)。结论SNHG17在支气管哮喘幼鼠模型ASMCs中上调,可通过miR-375促进ASMCs增殖和迁移。Objective To explore the expression and role of long non coding RNA(LncRNA)small nuclear RNA host gene 17(SNHG17)in airway smooth muscle cells(ASMCs).Methods The asthmatic model of young Wistar rats was constructed by intraperitoneal injection of ovalbumin antigen suspension.The rats were randomly divided into control group(injected intraperitoneally with normal saline)and model group(injected intraperitoneally with 10%ovalbumin antigen suspension),the blank plasmid(pc-NC group)and SNHG17 of overexpression plasmid(pc-SNHG17 group)were transfected into ASMCs of the model group,respectively.The expression levels of SNHG17 and miR-375 in ASMCs were detected by real-time quantitative polymerase chain reaction(qRT-PCR).The proliferation rates of ASMCs were detected by EdU assay,and the migration rates of ASMCs was detected by wound healing assay.Results The expression levels of SNHG17 in ASMCs of control group and model group were 1.00±0.10 and 1.81±0.11,respectively.The expression levels of miR-375 in ASMCs of control group,model group,pc-NC group and pc-SNHG17 group were1.01±0.08,0.72±0.05,0.79±0.05 and 0.53±0.06,respectively.The cell proliferation rates of control group,model group,pc-NC group and pc-SNHG17 group were(26.11±2.51)%,(33.01±2.91)%,(34.44±3.07)%,and(43.92±2.97)%,respectively.The cell migration rates of control group,model group,pc-NC group and pc-SNHG17 group were(10.05±1.00)%,(18.62±1.20)%,(20.03±1.12)%,and(44.71±3.33)%,respectively.All the above data showed that the differences between the model group and the control group,the pc-SNHG17 group between the pc-NC group,were statistically significant(all P<0.05).Conclusion SNHG17 was up-regulated in ASMCs of young asthma rat model,and could promote the proliferation and migration of airway smooth muscle cells through miR-375.
