摘 要:目的研究异佛司可林通过调控长链非编码RNA(lncRNA)Nkx2.1相关的非编码RNA(NANCI)表达对高氧诱导新生小鼠肺损伤的保护作用。方法将新生小鼠分成对照组、模型组(高氧诱导肺损伤)、实验组(高氧诱导肺损伤,20 mg·kg^(-1)的异佛司可林治疗)、实验+si-NC组(高氧诱导肺损伤,20 mg·kg^(-1)的异佛司可林+、siRNA control)、实验+si-NANCI组(高氧诱导肺损伤,20 mg·kg^(-1)的异佛司可林+NANCI siRNA治疗)。用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测NANCI表达,检测小鼠肺功能和肺湿干比值,用苏木精-伊红(HE)染色对肺损伤评分,用微量法检测超氧化物歧化酶(SOD)、丙二醛(MDA)水平,以酶联免疫吸附(ELISA)法检测白细胞介素-1β(IL-1β)水平。结果对照组、模型组、实验组、实验+si-NC组和实验+si-NANCI组新生小鼠肺组织中NANCI表达水平分别为1.00±0.06,0.28±0.03,0.75±0.09,0.75±0.06和0.33±0.02,静息通气量分别为(3.16±0.18),(1.23±0.12),(2.31±0.11),(2.29±0.16)和(1.79±0.10)mL·kg^(-1),肺容积分别为(0.26±0.02),(0.13±0.02),(0.26±0.01),(0.25±0.03)和(0.17±0.02)mL,肺损伤评分分别为2.15±0.16,8.27±0.33,3.16±0.32,3.07±0.23和6.43±0.49,肺湿干比值分别为3.24±0.16,7.11±0.67,3.87±0.25,3.77±0.40和5.23±0.44,肺组织中SOD含量分别为(333.80±20.00),(207.98±8.95),(321.96±14.47),(322.74±13.42)和(243.73±15.44)U·mg^(-1),MDA含量分别为(6.47±0.65),(12.09±1.02),(8.06±0.73),(7.96±0.81)和(9.32±1.22)nmol·mg^(-1),IL-1β含量分别为(77.17±6.06),(338.30±22.69),(81.14±4.26),(80.85±4.73)和(179.63±16.93)ng·g^(-1)。上述指标,模型组与对照组比较,差异均有统计学意义(均P<0.05);实验组与模型组比较,差异均有统计学意义(均P<0.05);实验+si-NANCI组与实验+si-NC组比较,差异均有统计学意义(均P<0.05)。结论异佛司可林可通过上调NANCI表达减轻高氧诱导新生小鼠肺损伤。Objective To investigate the protective effect of isoforskolin on the lung injury of neonatal mice induced by hyperoxia through regulating the expression of long noncoding RNA(lncRNA)Nkx2.1-associated non-codIng RNA(NANCI).Methods Neonatal mice were divided into control group,model group(hyperoxia-induced lung injury),experimental group(hyperoxia-induced lung injury,20 mg·kg^(-1) isoforskolin treatment),experimental+si-NC group(hyperoxia-induced lung injury,20 mg·kg^(-1) isoforskolin,siRNA control treatment),experimental+si-NANCI group(hyperoxia-induced lung injury,20 mg·kg^(-1)isoforskolin,NANCI siRNA treatment).Real-time fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)method was used to detect NANCI expression,mouse lung function and lung wet-dry ratio was detected,lung injury scores were evaluated by hematoxylin-eosin(HE)staining,superoxide dismutase(SOD)and malonaldehyde(MDA)were detected by microassay,interleukin-1β(IL-1β)levels were detected by enzyme-linked immunosorbent assay(ELISA).Results The expression levels of NANCI in the lung tissue of newborn mice in control group,model group,experimental group,experimental+si-NC group,experimental+si-NANCI group were 1.00±0.06,0.28±0.03,0.75±0.09,0.75±0.06,0.33±0.02;the resting ventilation volume were(3.16±0.18),(1.23±0.12),(2.31±0.11),(2.29±0.16),(1.79±0.10)m L·kg^(-1);lung volumes were(0.26±0.02),(0.13±0.02),(0.26±0.01),(0.25±0.03),(0.17±0.02)m L,lung injury scores were2.15±0.16,8.27±0.33,3.16±0.32,3.07±0.23,6.43±0.49,lung wet-to-dry ratios were 3.24±0.16,7.11±0.67,3.87±0.25,3.77±0.40,5.23±0.44;SOD content in lung tissue were(333.80±20.00),(207.98±8.95),(321.96±14.47),(322.74±13.42),(243.73±15.44)U·mg^(-1);MDA content were(6.47±0.65),(12.09±1.02),(8.06±0.73),(7.96±0.81),(9.32±1.22)nmol·mg^(-1);IL-1βcontent were(77.17±6.06),(338.30±22.69),(81.14±4.26),(80.85±4.73),(179.63±16.93)ng·g^(-1).Compared model group with control group,the difference of the above indicators were all stati
