摘 要:目的观察石墨烯膜材料联合米诺地尔对小鼠毛发生长的影响。方法2020年10月至2021年5月,对24只6周龄雄性C57BL/6j小鼠进行背部脱毛后,采用随机数字表法分为正常组、石墨烯膜组、米诺地尔组、实验组,每组6只。正常组仅进行了脱毛处理,石墨烯膜组于脱毛区域每天使用石墨烯膜材料覆盖并通电处理1 h,米诺地尔组每天外用5%米诺地尔1 ml,实验组每天外用5%米诺地尔1 ml后,再局部使用石墨烯膜材料通电处理1 h。干预2周,通过苏木精-伊红(HE)染色,毛囊细胞凋亡原位末端转移酶标记技术(TUNEL)免疫荧光染色,细胞增殖抗原(Ki-67)免疫荧光染色,实时免疫荧光聚合酶链反应法(qRT-PCR)检测Wnt5a蛋白(Wnt5a)、Wnt10b蛋白(Wnt10b)和β-连环蛋白(β-Catenin)的mRNA相对表达量,蛋白质印迹法(Western blot)检测β-catenin和Wnt10b的蛋白相对表达水平。多组间采用多因素分析。结果组织学结果显示,与其他3组比较,实验组小鼠的毛囊数目明显增多,毛乳头膨大,深入皮下组织,多数处于生长期。qRT-PCR实验结果显示,在处理第14天,实验组Wnt5a高于米诺地尔组(7.37±0.14比4.93±0.15,F=0.15,P<0.01),实验组Wnt10b高于米诺地尔组(7.92±0.08比6.45±0.09,F=0.14,P<0.01)、实验组β-catenin高于米诺地尔组(7.37±0.06比6.53±0.04,F=0.57,P<0.01)。蛋白质印迹法(Western blot)实验结果显示,实验组β-catenin表达水平高于米诺地尔组(1.48比1.17),实验组Wnt10b表达水平高于米诺地尔组(1.19比0.90)。结论石墨烯膜材料联合米诺地尔可能通过上调Wnt/β-catenin信号通路表达水平来调控小鼠的毛囊发育及毛发生长。Objective To observe the effect of graphene membrane material combined with minoxidil on hair growth in mice.Methods From October 2020 to May 2021,246-week-old male C57BL/6j mice were divided into normal group,graphene film group,minoxidil group,and experimental group by random number table method after back depilation.The normal group only underwent hair removal treatment.The graphene film group was covered with graphene film material and energized for 1 h every day in the hair removal area.The minoxidil group was treated with 5%minoxidil 1 ml every day,and the experimental group was treated with 5%minoxidil every day and the graphene membrane material was used locally for 1 h of electrification.After 2 weeks of intervention,hematoxylin-eosin(HE)staining,terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling(TUNEL)immunofluorescence staining,cell proliferation antigen(Ki-67)immunofluorescence staining and quantitative real-time polymerase chain reaction(qRT-PCR)were used to detect the relative mRNA expression of Wnt5a,Wnt10b andβ-catenin.The relative protein expression levels ofβ-catenin and Wnt10b were detected by Western blotting.Multivariate analysis was used between multiple groups.Results The histological results showed that as compared with the other three groups,the number of hair follicles in the experimental group was significantly increased,and the dermal papillae were enlarged and penetrated into the subcutaneous tissue,most of which were in the growth phase.The results of qRT-PCR experiment showed that on the 14th day of treatment,Wnt5a in the experimental group was higher than that in the minoxidil group(7.37±0.14 vs.4.93±0.15,F=0.15,P<0.01),and Wnt10b in the experimental group was higher than that in the minoxidil group(7.92±0.08 vs.6.45±0.09,F=0.14,P<0.01),theβ-catenin in the experimental group was higher than that in the minoxidil group(7.37±0.06 vs.6.53±0.04,F=0.57,P<0.01).The results of Western blotting experiment showed that the expression level ofβ-catenin in the expe
