摘 要:目的探讨白屈菜碱对乳腺癌MCF-7细胞凋亡调控的作用及机制。方法分别以白屈菜碱和二甲基亚砜处理MCF-7细胞作为实验组和对照组。以细胞计数试剂-8(CCK-8)法检测细胞活性;通过蛋白质印迹法(Western blot)检测p65、B细胞淋巴瘤/白血病-2(bcl-2)、bcl-2相关X蛋白(bax)表达;通过流式细胞术检测细胞凋亡。应用GraphPad Prism 9统计分析,组间比较采用Student’s t检验分析。结果CCK-8显示实验组A450(0.414±0.023)小于对照组A450(1.042±0.190),差异有统计学意义(t=5.683,P<0.01);流式细胞术结果显示,实验组细胞凋亡率[(20.14±3.25)%]高于对照组[(4.12±0.53)%],差异有统计学意义(t=8.426,P<0.01);Western blot结果显示,总蛋白实验组p65表达量(1.526±0.251)与对照组(1.624±0.208),差异无统计学意义(t=0.521,P>0.05),bcl-2表达量(0.486±0.063)低于对照组(0.785±0.098),差异有统计学意义(t=4.445,P<0.05),bax表达量(1.235±0.251)高于对照组(0.426±0.015),差异有统计学意义(t=5.572,P<0.01);对核蛋白和胞质蛋白进行分别检测,结果提示实验组细胞核内p65表达量(0.565±0.075)低于对照组(0.956±0.065),差异有统计学意义(t=6.823,P<0.01),而实验组胞质中p65表达量(0.725±0.125)高于于对照组(0.327±0.089),差异有统计学意义(t=0.492,P<0.05)。结论白屈菜碱通过抑制p65核转位抑制核因子-κB信号促进MCF-7细胞凋亡。Objective To investigate the effect and mechanism of chelidonine on apoptosis of MCF-7 breast cancer cells.Methods MCF-7 cells treated with chelidonine and dimethyl sulfoxide(DMSO)were used as experimental group and control group respectively.Cell viability was determined by cell counting kit-8(CCK-8)assay.The expression of p65,B cell lymphoma/leukemia-2(bcl-2)and bcl-2 associated X protein(bax)was detected by Western blotting.Apoptosis rate was detected by flow cytometry.GraphPad Prism 9 was used for statistical analysis,and Student’s t test was used for group comparison.Results The CCK-8 assay showed that the A450 in the experimental group(0.414±0.023)was lower than that in the control group(1.042±0.19,t=5.683,P<0.01).The flow cytometry results showed that the apoptosis rate of the experimental group[(20.14±3.25)%]was higher than that of the control group[(4.12±0.53)%,t=8.426,P<0.01].Western blotting showed that there was no significant difference in the p65 expression between the experimental group(1.526±0.251)and the control group(1.624±0.208,t=0.521,P<0.05).The expression level of Bcl-2(0.486±0.063)was significantly lower in the experimental group than that in the control group(0.785±0.098,t=4.445,P<0.05).The expression level of bax(1.235±0.251)was significantly higher in the experimental group than that in the control group(0.426±0.015,t=5.572,P<0.01).Nuclear protein and cytoplasmic protein were detected,and the results showed that the expression of p65 in the nuclei of the experimental group(0.565±0.075)was significantly lower in the experimental group than that in the control group(0.956±0.065,t=6.823,P<0.01).The expression level of p65 in the experimental group(0.725±0.125)was significantly higher than that in the control group(0.327±0.089,t=0.492,P<0.05).Conclusion Chelidonine can promote apoptosis of MCF-7 cells by inhibiting nuclear translocation of p65 and inhibiting nuclear factor-κB signal.
