摘  要:目的探究一种新型可注射型水凝胶早期促进骨髓间充质干细胞向成骨细胞分化的能力,以及在体内修复大鼠颅骨缺损的效果和可行性。方法使用丝素蛋白溶液和磷酸镁基水凝胶制备成不同磷酸镁含量的4组水凝胶,分别命名为Mg0、Mg1、Mg2和Mg5并研究分析材料表征。体外实验检测水凝胶的生物活性及诱导成骨分化的能力。体内实验采用10只SD大鼠进行颅骨缺损模型制备,简单随机法分为实验组和空白组,对该种新型可注射型水凝胶用于修复缺损颅骨的疗效及可行性进行评价。计量资料应用Graph Pad 9.0进行ANOVA方差分析。结果电镜扫描(SEM)显示了4组水凝胶的表面微观形态。细胞计数试剂盒(CCK-8)结果示共培养1 d时对照组与Mg0、Mg1、Mg2和Mg5的吸光度(A)值分别为0.399±0.024、0.436±0.043、0.364±0.020、0.3205±0.029和0.338±0.028,在共培养3 d时为0.606±0.045、0.590±0.042、0.557±0.014、0.453±0.056和0.610±0.046,第5天时分别为0.882±0.022、0.841±0.147、0.824±0.040、0.828±0.049和0.830±0.021,第7天时分别为1.162±0.029、1.397±0.056、1.059±0.050、1.063±0.084和1.102±0.046(F(12,77)=8.0,P<0.01)。使用氯化十六烷吡啶(CPC)对14 d时的成骨矿化物进行定量,Mg0、Mg1、Mg2和Mg5的A值结果分别为0.937±0.107、1.646±0.507、2.181±0.038及2.006±0.043(F=56.0,P<0.05)。体内实验示可注射型水凝胶修复效果显著,在修复后第4周可见大量新生骨胶原。结论丝素蛋白-磷酸镁基可注射型水凝胶能有效修复骨缺损,并可在体内促进早期骨胶原的形成。Objective A new type of injectable hydrogel for bone repair was prepared to analyze its osteogenic ability in vitro and in vivo.Methods A total of four injectable hydrogels were prepared by silk fibroin and magnesium phosphate-based gel in different ratios and termed as Mg0,Mg1,Mg2 and Mg5,respectively.Morphology analyses were firstly carried out before an in vitro study for bioactivity and osteogenic ability.Then cranial defects were created on ten 8-week-old male rats and randomly divided into an SF-Mg group and a control group.The two groups were treated with and without hydrogel implantation to furtherly confirm the validity and feasibility of the injectable hydrogel.All data collected were analyzed by GraphPad 9.0.Results The micromorphological changes on the surface of 4 injectable hydrogels were shown under scanning electrical microscope.Cell counting kit-8(CCK-8)results showed the absorbance(A)values in control group,Mg0,Mg1,Mg2 and Mg5 were 0.399±0.024,0.436±0.043,0.364±0.020,0.3205±0.029 and 0.338±0.028 on day 1,0.606±0.045,0.590±0.042,0.557±0.014,0.453±0.056 and 0.610±0.046 on day 3,0.882±0.022,0.841±0.147,0.824±0.040,0.828±0.049 and 0.830±0.021 on day 5,1.162±0.029,1.397±0.056,1.059±0.050,1.063±0.084 and 1.102±0.046 on day 7,respectively[F(12,77)=8.0,P<0.01].The CPC analysis for mineralization on day 14 showed that the values in Mg0,Mg1,Mg2 and Mg5 were 0.937±0.107,1.646±0.507,2.181±0.038 and 2.006±0.043(respectively,F=56.0,P<0.05).Conclusion The silk fibroin-magnesium phosphate injectable hydrogel can be applied for bone repair with osteogenic ability in early stage.
