摘 要:目的 观察大气污染慢性暴露对健康大鼠喉黏膜中细胞因子水平的影响。方法 12只健康SD大鼠随机分成2组,暴露组置于染毒仓内接受北京市2016年12月17日~2017年2月17日实时大气污染暴露,对照组置于SPF级环境中进行标准化饲养。完成暴露后取材喉黏膜组织,HE染色观察病理形态学变化,液相芯片检测促炎细胞因子,包括白细胞介素1β(IL-1β)、IL-18、肿瘤坏死因子α(TNF-α)、IL-17A,以及抗炎细胞因子,包括IL-4、IL-10、IL-13的表达水平变化。结果 经慢性暴露后,喉黏膜上皮角蛋白层增厚、细胞排列紊乱、基底部细胞增生、组织间质增宽和中性粒细胞增多。促炎性细胞因子IL-1β、IL-18和IL-17A和抗炎细胞因子IL-13表达显著升高(P<0.05)。结论 大气污染慢性暴露通过诱导喉黏膜局部促炎/抗炎细胞因子表达失衡,发挥炎症损伤作用。OBJECTIVE To observe the effect of chronic exposure to air pollution on cytokines in laryngeal mucosa of healthy rats.METHODS Twelve healthy SD rats were randomly divided into two groups.The exposure group was exposed to real-time air pollution in Beijing from December 2016 to February 2017,and the control group was placed in SPF environment for standardized feeding.The laryngeal mucosa tissues were taken after exposure.The histopathological changes were observed by HE staining,and the expression levels of pro-inflammatory cytokines(IL-1β,IL-18,TNF-α,IL-17A) and anti-inflammatory cytokines(IL-4,IL-10,IL-13) were detected by liquid chip.RESULTSAfter chronic exposure,the epithelial keratin layer of laryngeal mucosa was thickened,the cell arrangement was disordered,the basal cells were proliferated,the interstitial tissue was widened,and neutrophils were increased.The expression of pro-inflammatory cytokines IL-1β,IL-18 and IL-17A and anti-inflammatory cytokines IL-13 were significantly increased(P<0.05).CONCLUSION Chronic exposure to air pollution plays an inflammatory injury role by inducing the imbalance of expression of pro-inflammatory/anti-inflammatory cytokines in laryngeal mucosa.
