摘 要:目的:探讨降糖益肾丸的抗糖尿病肾病(DN)作用。方法:SPF级雄性SD大鼠随机分为对照组与DN模型组,正常对照组给予普通饲料喂养,其余DN模型组大鼠采用高脂高糖饮食联合链脲佐菌素(STZ)注射建立DN模型,按DN模型+降糖益肾丸组(高剂量组)、DN模型+降糖益肾丸组(中剂量组)、DN模型+降糖益肾丸组(低剂量组)、DN模型+盐酸二甲双胍+厄贝沙坦组(阳性药组),持续4周给予相应药物;动物处理并收集样品,分别测定各组大鼠空腹血糖(FBG)、24h尿蛋白(24h-Pro)、血肌酐(Scr)及血尿素氮(BUN)的含量;采用苏木素-伊红(HE)染色观察各组大鼠肾组织的形态结构变化。结果:与对照组比较,模型组大鼠FBG、24h-Pro、Scr、BUN的含量均显著升高(P<0.01)。与模型组比较,阳性药组与中、高剂量组大鼠FBG、24h-Pro、Scr、BUN的含量均显著降低,低剂量组大鼠FBG、24h-Pro、Scr、BUN的含量亦下降(P<0.01或P<0.05)。与中、高剂量组比较,低剂量组FBG、24 h-Pro、Scr、BUN的含量显著升高(P<0.01);正常对照组大鼠肾细胞形态结构正常。与正常对照组比较,模型组肾小球体积明显增大,出现较多的空泡,上皮细胞脱落至管腔,皮质区肾小管有扩张合并空泡变,表明肾小球存在一定程度的损伤。与模型组比较,各药物组大鼠肾小球大多数体积、空泡变小,肾小球受损程度有所缓解。结论:降糖益肾丸对模型大鼠具有明显的抗DN肾损伤作用。Objective:To investigate the anti-diabetic nephropathy(DN)effect of Jiangtang Yishen Pills.Methods:SPF male SD rats were randomly divided into the control group and the DN model group.The normal control group was fed with ordinary feed.The other DN model groups were fed with high-fat and high-sugar diet combined with streptozotocin(STZ)injection to establish the DN model.The DN model+Jiangtang Yishen pills group(high dose group),DN model+Jiangtang Yishen pills group(middle dose group),DN model+Jiangtang Yishen pills group(low dose group),DN model+metformin hydrochloride+irbesartan group(positive drug group)were given the corresponding drugs for four weeks.The animals were treated and the samples were collected,and the contents of fasting blood glucose(FBG),24hurinary protein(24h-Pro),serum creatinine(Scr)and blood urea nitrogen(BUN)in each group were measured.hematoxylin-eosin(HE)staining was used to observe the morphological changes of renal tissue in rats in each group.Results:Compared with the control group,the contents of FBG,24h-Pro,Scr and BUN in the model group were significantly increased(P<0.01).Compared with the model group,the contents of FBG,24h-Pro,Scr and BUN in the positive drug group and the middle and high dose groups were significantly decreased,and the contents of FBG,24hPro,Scr and BUN in the low dose group were also decreased(P<0.01or P<0.05).Compared with the middle and high dose groups,the contents of FBG,24h-Pro,Scr and BUN in the low dose group were significantly increased(P<0.01).The morphological structure of renal cells in normal control group was normal.Compared with the normal control group,the glomerular volume of the model group was significantly increased,and there were more vacuoles.The epithelial cells fell off to the lumen,and the renal tubules in the cortex were dilated and vacuolated,indicating that there was a certain degree of glomerular injury.Compared with the model group,most of the glomerular volumes and vacuoles of rats in each drug group were smaller,and the glomerula
