摘  要:目的:观察绞股蓝皂苷对慢性SD大鼠高眼压模型中视网膜STAT3和JAK2mRNA表达的影响。方法:在60只大鼠中随机抽取10只作为空白组,余下50只大鼠建立为慢性高眼压模型。造模成功后的50只大鼠均给与噻马洛尔滴眼液降眼压治疗,并随机分为5组,分别为阴性对照组:予以生理盐水灌胃;低剂量组:绞股蓝皂苷25mg/(kg·d)灌胃;中剂量组:绞股蓝皂苷50mg/(kg·d)灌胃;高剂量组:绞股蓝皂苷100mg/(kg·d)灌胃;阳性对照组:维生素B1和维生素B12灌胃治疗,每组各10只大鼠。造模后的第1天至第14天(D1-D14),对大鼠术眼进行跟踪观察。灌胃至第14天并将大鼠处死,进行实时PCR含量测定。结果:qPCR结果显示,空白组、绞股蓝各治疗组和阳性对照组STAT3mRNA和JAK2mRNA的相对表达量明显低于阴性对照组(P<0.05),高剂量组低于中剂量组(P<0.05)。结论:绞股蓝皂甙可以抑制STAT3mRNA和JAK2mRNA的表达,对慢性高眼压大鼠视网膜神经节细胞有一定保护作用。Objective:To observe the protective effect of gynostemma glycosides on the mRNA expression of retinal STAT3and JAK2in a rats with chronically high intraocular pressure.Methods:A total of 60rats were randomly divided in group A(the blank group,10rats)and a chronic high IOP model group(50rats).The IOP model group(IOP above 22mmHg)was then randomly divided into an additional 5groups(10rats per group):group B(negative control group)treated with normal saline;group C treated with gynostemma glycosides 25mg/(kg·d);group D treated with gynostemma glycosides 50mg/(kg·d);group E treated with gynostemma glycosides 100mg/(kg·d);group F(positive control group)treated with VitB1and VitB12.The eyes of each rat were monitored from day 1to 14(D1-D14).On day 14,rats were euthanized,after which retinal tissue and optic nerve were examined using real-time PCR.Results:The relative expression of STAT3mRNA and JAK2(mRNA and protein)in groups A,C,D,E,and F was significantly lower than in group B(P<0.05),while in group E,the expression was lower than in group D(P<0.05).Conclusion:Gynostemma glycosides can effectively inhibit the expression of TAT3/JAK2mRNA to protect the optic nerve of rats with chronic high intraocular pressure.
