摘 要:目的:研究光皮木瓜的薄层色谱,为光皮木瓜对照药材标定标准的建立提供参考。方法:在《湖北省中药材质量标准》2018年版光皮木瓜质量标准基础上进行改进,采用聚酰胺薄膜与硅胶薄层色谱鉴别法,新增槲皮苷、芦丁、绿原酸、熊果酸作为对照物质,对不同产地的光皮木瓜样品进行研究。结果:以聚酰胺薄膜为吸附剂,甲醇-冰醋酸-水(4∶1∶5)为展开剂,较好分离了光皮木瓜中多个黄酮类色谱斑点,同时指认了色谱中的槲皮苷、芦丁、绿原酸;采用硅胶薄层色谱鉴别了以熊果酸为代表的三萜酸。不同产地光皮木瓜的薄层色谱基本一致,且与易混品木瓜有明显区别。结论:改进后的光皮木瓜薄层鉴别方法的分离度、准确性提高,重复性好,为光皮木瓜对照药材标定标准的薄层鉴别方法的建立提供了科学依据。Objective:To study Thin-layer chromatography(TLC)of Chaenomelis Sinensis Fructus,in order to provide basis for establishment of calibration standard for reference drug of Chaenomelis Sinensis Fructus.Methods:TLC identification Method was improved on the basis of the Quality Standard of Chinese Crude Drugs in Hubei Province(2018edition).Polyamide film chromatography and silica gel TLC were applied.Quercitrin,Rutin,Chlorogenic acid and Ursolic acid were added as new standards.Chaenomelis Sinensis Fructus samples from different areas were analysed by the improved method.Results:A set of flavonoids sports on chromatograms of Chaenomelis Sinensis Fructus samples were fairly seperated by using the mobile phase methanol-acetic acid-water(4∶1∶5)on the polyamide film.Quercitrin,Rutin and Chlorogenic acid were recognized on the chromatograms.Triterpenic acid represented by Ursolic acid was identified by silica gel TLC.The chromatograms of Chaenomelis Sinensis Fructus from different producing areas were basically the same,and was obviously different from the adulterant Chaenomelis Fructus.Conclusion:The improved method has high resolution and accuracy,aslo reproducible,which can provide scientific basis for establishment of TLC identification methods of calibration standard for reference drug of Chaenomelis Sinensis Fructus.
