摘 要:目的探讨NUS1在宫腔粘连患者和正常子宫内膜组织中的表达情况,并分析其生物学功能。方法采用RT-qPCR和Western blot方法分析NUS1 mRNA及其蛋白在两组中的表达;End1/E6E7细胞转染质粒成功后,采用流式细胞实验来评估不同NUS1质粒对细胞周期和凋亡功能的影响。结果病例组NUS1 mRNA(2.66±0.16)及蛋白(2.49±0.03)的表达均明显高于对照组(P<0.05)。与对照组比较,NUS1过表达组细胞G期向S期转换,凋亡率明显下降,而敲降质粒组结果与之相反(0.89±0.01,1.08±0.03),差异有统计学意义(P<0.05)。结论NUS1在人宫腔粘连组织中异常高表达;NUS1可能参与End1/E6E7细胞体外凋亡能力。Objective Study the expression of NUS1 in patients with intrauterine adhesions and normal endometrial tissues,and explore its biological function.Methods Expressions of mRNA and protein of NUS1 in the two groups were analyzed by RT-QPCR and Western blot.After End1/E6 E7 cells were successfully transfected with plasmids,flow cytometry was used to evaluate the effects of different NUS1 plasmids on cell cycle and apoptosis.Results The mRNA(2.66±0.16)and protein(2.49±0.03)expressions of NUS1 in the case group were significantly higher than those in the control group(P<0.05).Compared to the control group,NUS1 overexpression group transformed from G phase to S phase,and the apoptosis rate decreased significantly,while the result of knockdown plasmids group was the opposite(0.89±0.01,1.08±0.03),The difference was statistically significant(P<0.05).Conclusion NUS1 is abnormally higher in human intrauterine adhesions;NUS1 may be involved in End1/E6 E7 cell apoptosis in vitro.
