摘 要:基于网络药理学探讨当归-红花治疗原发性痛经的潜在作用机制。利用TCMSP、SwissTargetPrediction、BindingDB、PharmMapper和BATMAN-TCM数据库获取当归-红花化学成分的作用靶点,使用GeneCards、NCBI、TTD、CTD和OMIM数据库获取原发性痛经疾病靶点,将药物与疾病交集靶点导入Cytoscape 3.7.0软件建立药物—化学成分—靶点—疾病网络,导入STRING数据库获取靶点PPI网络,导入R语言中Bioconductor的生物信息软件包进行生物功能注释(GO)和KEGG通路富集分析。结果显示:当归-红花中槲皮素、山奈酚、熊果酸、野黄芩素、咖啡酸、七叶亭、阿魏酸、东莨菪内酯、阿魏酸松柏酯和芦丁等24个成分,可能通过与炎症、激素调节、中枢镇痛、平滑肌解痉以及免疫相关的27条通路,干预PTGS2、CA2、PTGS1、AKR1B1、CA3、ESR2、ALOX5、TTR、MMP2以及EGFR等81个靶点,发挥治疗原发性痛经的作用。当归-红花治疗原发性痛经具有多成分、多靶点和多途径的特点,研究结果初步揭示了当归-红花治疗原发性痛经的潜在活性成分及可能的作用机制,为进一步的药效物质基础和作用机制实验研究提供了理论依据。To study the mechanism of herb pair containing Angelica sinensis radix and Carthami flos(ASR-CF) for treatment of primary dysmenorrhea based on network pharmacology.TCMSP,SwissTargetPrediction,BindingDB,PharmMapper and BATMAN-TCM were used to obtain the target of the components in ASR-CF.GeneCards,NCBI,TTD,CTD and OMIM were used to obtain the target of primary dysmenorrhea.Cytoscape 3.7.0 software was used to create a drug-chemical component-target-disease network.Using STRING to establish and analyze the PPI network,and Bioconductor was used to analyze the GO function and KEGG pathway enrichment of the targets.Results show that 24 effective compounds of ASR-CF including quercetin,kaempferol,ursolic acid,scutellarin,caffeic acid,esculetin,ferulic acid,scopolactone,coniferulate,rutin might act on 81 targets including PTGS2,CA2,PTGS1,AKR1 B1,CA3,ESR2,ALOX5,TTR,MMP2,EGFR through 27 pathways related to inflammation,hormonal regulation,central analgesia,smooth muscle spasmolysis and immunity for treatment of primary dysmenorrhea.ASC-CF had the characteristics of multi-component,multi-target and multi-channel in the treatment of primary dysmenorrhea,and the results reveal the active ingredients and potential mechanism of ASC-CF in the treatment of primary dysmenorrhea.Our findings provide a theoretical basis for further experimental study on the material basis and mechanism of action.
